Untargeted metabolomics links glutathione to bacterial cell cycle progression

Abstract: Cell cycle progression requires the coordination of cell growth, chromosome replication, and division. Consequently, a functional cell cycle must be coupled with metabolism. However, direct measurements of metabolome dynamics remained scarce, in particular in bacteria. Here, we describe an untargeted metabolomics approach with synchronized Caulobacter crescentus cells to monitor the relative abundance changes of ~400 putative metabolites as a function of the cell cycle. While the majorit… Show more

“…Notably, tedious sample pretreatment (e.g., derivatization) and lengthy chromatographic separation (≈40 min) are required to reduce biosample complexity and enrich target molecules in conventional MS, including liquid chromatography‐mass spectrometry and gas chromatography‐mass spectrometry. [ 28 ] In contrast to the conventional MS, our approach avoided the sample treatment procedure, suitable for large scale clinical use with enhanced analytical speed (approximately seconds), reduced sample consumption (down to ≈nL), and low cost (≈€3 per test). “…Global or untargeted metabolomics is increasingly used to investigate metabolic changes of various biological or environmental systems in an unbiased manner [1,2]. Liquid chromatography coupled to high-resolution mass spectrometry (LC-HRMS) has become the main workhorse for global metabolomics [3,4].…”

“…Glutathione can reduce oxidized glutaredoxins, which serve as antioxidants. Both glutathione and thioredoxin have been shown to play a role in cell cycle regulation, and their concentrations vary between different growth phases [ 62 , 63 ]. However, Cu can also form a catalytic complex with glutathione, generating additional ROS instead of depleting them [ 64 ].…”
