In the carrot tissue culture system used the transition from log-Phase to stationary-Phase of cell division activity is usually associated with root formation. In -Kinetin cultures. this transition as well as root formation occurs after 14-17 days in cultures, whereas in +Kinetin treatments this was delayed to 4-5 weeks o f culture. In the presence and absence of Kinetin, R N A content, protein content, amino acid composition of protein as well as the activity of some enzymes (Aldolase and Fumarase) was followed during a culture period of 28 days. O n a cellular basis RNA and protein content in -Kinetin tissues was throughout higher than in + Kinetin treatments. The activity of aldolase and fumarase followed protein content in the beginning of the culture period.From all these results, a culture period of 12 and 28 days seemed to be o f particular interest. A comparison of the pattern of analytical disc-electropherograms o f soluble protein as well as enzyme activity (Aldolase, Fumarase, Amylase, Glutamate-Oxalacetat-Transaminase, Glutamate-Pyruvate-Transaminase, RNase and proteolytic, activity) of carrot tissues after these periods of culture ( k Kinetin) with that of the original tissues revealed characteristic changes dtte to Kinetin as .well as to the period of culture. Furthermore, the activity of some of these enzymes is compared with corresponding metabolic data.
