Up‐regulation of estrogen receptor‐β expression during osteogenic differentiation of human periodontal ligament cells

Abstract: The results suggest that estrogen receptor-beta may be the predominant subtype expressed in human periodontal ligament cells and may actively participate in the osteogenic differentiation process of human periodontal ligament cells, both in male and in female subjects.

“…As has been reported in other cell types, expression of both ERα and ERβ were observed to increase overtime in osteogenic medium, (Fig. 4A, N = 3, * P <0.01) [37], [40]. After 7d differentiation, staining and quantification of alkaline phosphatase (ALP) activity was performed as an early marker of osteogenic differentiation (Fig.…”

“…Primary culture of hGF and hPDLC was carried out according to previously described methods [7], [26], [36], [43]. Briefly, hPDLC were prepared from extracted third molars of 5 young healthy volunteers and hGF were obtained from gingiva of the same 5 donors.…”

“…The remaining cells were harvested using lysis buffer [20 mM Tris (pH 7.4), 150 mM NaCl, 1 mM EDTA, 1 mM EGTA, 1% (v/v) Triton X-100, 2.5 mM sodium pyrophosphate, 1 mM β-glycerol phosphate and 2 mM Na 3 VO 4 supplemented with protease inhibitor cocktail (Roche, Mannheim, Germany)] [43] for Western blotting. Protein concentration was determined using the Bicinchoninic Acid Protein Assay Kit (Applygen, Beijing, China).…”
