Upregulated Expression of Activation-Induced Cytidine Deaminase in Ocular Adnexal Marginal Zone Lymphoma with IgG4-Positive Cells

Abstract: Immunoglobulin G4-related disease (IgG4-RD) is a systemic disorder characterized by tissue fibrosis and intense lymphoplasmacytic infiltration, causing progressive organ dysfunction. Activation-induced cytidine deaminase (AID), a deaminase normally expressed in activated B-cells in germinal centers, edits ribonucleotides to induce somatic hypermutation and class switching of immunoglobulin. While AID expression is strictly controlled under physiological conditions, chronic inflammation has been noted to induce… Show more

“…It may correlate with MZL with surrounding polyclonal IgG4‐positive cells, and IgG4‐producing MZL (Figure 2). As previously reported, activation‐induced cytidine deaminase overexpression, a potential oncogenic driver also noted in double‐hit lymphoma, could be mediating the oncogenesis in patients with IgG4‐RD to IgG4‐positive MZLs 11 . In de novo IgG4‐positive MZLs, IgG4 itself secreted by clonal IgG4‐producing tumor cells could be a driver of tumorigenesis.…”

“…In particular, marginal zone lymphoma (MZL), a type of non‐Hodgkin lymphoma, has been reported to arise in the context of IgG4‐RD in various organs such as the ocular adnexa 10,11 . Although it is rare, pathological analyses of ocular MZLs showed clonal IgG4‐positive or IgG4‐producing lymphoma cells forming a unique disease entity 12,13 .…”

“…Infraorbital nerve enlargement is a unique feature on MRI scans [ 31 ]. Upregulation of activation-induced cytidine deaminase (AID)—the master factor for somatic hypermutation and class-switch recombination of immunoglobulin genes—was observed in IgG4-related ophthalmic disease and OAMZL, whereas AID expression was lower in IgG4-negative OAMZL [ 36 ]. AID might be a driver for oncogenesis in the development of IgG4-related ophthalmic disease to IgG4-positive OAMZL.…”
