Upregulation of αvβ6 integrin, a potent TGF-β1 activator, and posterior capsule opacification

“…During the embryonic development of eyes, LEC proliferation and differentiation are affected by various factors outside the cells, in which TGF-β is one of the most important trans-differentiation cytokines (de Iongh et al, 2005). More and more studies confirm that TGF-β can lead to the apoptosis of LECs and induce cataract formation (Sponer et al, 2005). Under physiological conditions, TGF-β 2 is in the inactive form, and activity can be induced by some pathological changes such as surgery and trauma (Saika et al, 2005;Ou et al, 2009).…”

“…Injury by mechanical trauma can modulate integrin expression, which has particular relevance to fibrotic conditions that occur following surgery such as PCO. An interesting study was carried out by Sponer et al [144] using the human capsular bag system which demonstrated that aVb6 integrin is increased in capsular bags cultured in protein-free medium compared with cultured intact lenses. Similarly, in non-ocular fibrotic diseases aVb6 integrin is increased following acute biliary injury [145].…”

“…While many β subunits can form heterodimers with αv, in this study the increased expression of αv in response to TGFβ is paralleled only by upregulation of the αv heterodimer partner β5 (Dawes et al, 2007). However, in a study using the human lens capsular bag model for PCO, mock cataract surgery induces expression of αvβ6 integrin (Sponer et al, 2005). It is not surprising that distinct αv integrin receptors are induced in different models of lens fibrotic disease, as most αv integrin receptors are strongly implicated in the fibrotic disease process, particularly through their activation of latent forms of TGFβ (Sheppard, 2004) and their regulation of TGFβ receptor function.…”
