Abstract: While balafilcon A released the most drug from the silicone hydrogel materials, all materials released the drug too quickly to be effective as drug delivery devices.

“…In comparison, for ketotifen fumarate (0.2 mg/mL) and ketorolac tromethamine (0.3 mg/mL), the maximum uptakes were 100 to 230 and 60 to 125 g/lens, respectively, resulting in percentage releases of 48% to 65% and 1.5% to 28%, respectively. 43 In a more recent study on ciprofloxacin from our group, 49 which included new generation silicone hydrogel materials and used a slightly modified method to the previous ciprofloxacin study, 42 we were able to show that drug release varied from 0.016 Ϯ 0.004 mg/lens for lotrafilcon A lenses to 0.42 Ϯ 0.03 mg/lens for etafilcon A lenses, with an average of 0.133 mg/lens.…”

“…In the next decade, increasing numbers of children, teenagers, and young adults around the world will receive contact lenses for both cosmetic and medical purposes (myopia control 10Y17 , drug delivery 18,19 ). With the trend toward fitting SCLs to patients at a younger age, it will be even more important to understand the relationship between SCL-related behaviors, patterns of use, environmental exposures, and health outcomes with SCL wear.…”

“…17,19,20 Furthermore, previous in vitro studies only provided an understanding of drug release kinetics. 18,[21][22][23][24] In developing a drug delivery device for ocular mycoses, it is also important to show that these CLs are effective in inhibiting microbial growth. Thus, in this study, an agar-based eye model was developed and used to evaluate the growth of C. albicans when exposed to an antifungal containing CL.…”

“…17,19,20 The lack of in vivo studies can be attributed to current in vitro models used to evaluate drug release from CLs, which are rudimentary and fail to adequately simulate the ocular environment. For instance, the typical in vitro model consists of testing drug release from a CL in a 2 to 5 mL vial, 18,[21][22][23][24] which neither mimics the on-eye tear volume nor the tear flow. Furthermore, measuring the quantity of drugs released from a CL over time does not necessarily correlate with the inhibition or killing of microbes.…”
