Uptake of oxytetracycline and its phytotoxicity to alfalfa (Medicago sativa L.)

Abstract: Plant uptake of antibiotic oxytetracycline is energy-dependent. AbstractA series of experiments were conducted in a hydroponic system to investigate the uptake of oxytetracycline (OTC) and its toxicity to alfalfa (Medicago sativa L.). OTC inhibited alfalfa shoot and root growth by up to 61% and 85%, respectively. The kinetics of OTC uptake could be well described by MichaeliseMenten equation with V max of 2.25 mmol g À1 fresh weight h À1 , and K m of 0.036 mM. The uptake of OTC by alfalfa was strongly inhibite… Show more

“…To further distinguish active transport from facilitated diffusion, metabolic inhibitors were employed to affect active uptake by disabling cellular energy production. 2,4-dinitrophenol (2,4-DNP) is considered primarily as metabolic inhibitor (Kong et al 2007;Zhan et al 2010;Wang et al 2011). It is regarded as an uncoupler of oxidative phosphorylation, and of proton-coupled fluxes at the plasma membrane and endomembrane via the dissipation of trans-membrane electrochemical gradients of protons (Kong et al 2007).…”

“…Both glycerol and silver are widely used to assay aquaporin activity (Kong et al 2007), but their inhibitory mechanisms differ greatly. Silver may react with the sulfhydryl group of a cysteine in the vicinity of the conserved NPA motif, thus effectively blocks the constriction region of the water channel.…”

“…Few prior studies have investigated wastewater irrigation as a continuous exposure route for the co-uptake of antimicrobials and pathogens into food crops. Similarly, prior studies of antimicrobial uptake were performed in hydroponic systems (Herklotz et al, 2010, Kong et al, 2007, Migliore et al, 2003and Wu et al, 2012 or by introducing antimicrobials via soil amendment (Boxall et al, 2006, Grote et al, 2007, Kumar et al, 2005, Sabourin et al, 2012and Seo et al, 2010.…”

“…OTC can significantly affect soil microbial activity and high concentrations can produce visual symptoms in plants including light green or yellow colours in the leaves of alfalfa (Boleas et al 2005;Kong et al 2007). Application of OTC has been shown to activate existing soil pollutants such as heavy metals in contaminated soils in previous studies in our laboratory, indicating an elevated environmental risk in soils with multiple contaminants (Chen et al 2014).…”
