Use of a group-reactive and other monoclonal antibodies in an enzyme immunodot assay for identification and presumptive serotyping of aquatic birnaviruses

Abstract: A panel of monoclonal antibodies (MAbs) was prepared and used to develop an enzyme immunodot assay for the rapid identification and presumptive serotyping of aquatic birnaviruses. Comparison of the reaction patterns of these MAbs with representative virus isolates indicated that one MAb recognizes a serogroupreactive epitope and can therefore be used for identification of all serogroup A aquatic birnaviruses, the predominant serotype worldwide. Other MAbs exhibited more restrictive specificities, permitting th… Show more

“…By cross neutralization tests with polyelonal antisera, aquatic birnaviruses are divided into two serogroups, A and B. Serogroup A comprises the nine serotypes WB, Sp, Ab, Ja, Te, He, Cl, C2 and C3. Serogroup B comprises a few unrelated isolates (Caswell- Reno, Lipipun, Reno & Nicholson 1989).…”

“…Traditionally, serotyping of IPNV is performed by cross neutralization tests with immune serum. More rapid immunological methods arc available (Nicholson & Hcnchal 1978;Piper, Nicholson & Dunn 1973;Tu, Spendlovc & Goede 1974;J0rgensen 1974;Hattori, Kodama, Ishiguro, Honda, Mikami & Izawa 1984;Dixon & Hill 1983;Rodak, Pospisil, Tomanek, Vesely, Obr & Valicek 1988;Nicholson & Caswell 1982), and recently methods for serotyping of IPNV by use of monoclonal antibodies have been reported (Caswell-Reno, Lipipun, Reno & Nicholson 1989;.…”

“…Six other serotypes were proposed by Hill & Way (1983 including four Canadian strains; Canada-1 (A6), Canada-2 (A7), Canada-3 (A8) and Jasper (A9). Caswell-Reno, Lipipun, Reno & Nicholson (1989) confirmed these additional serotypes using monoclonal antibodies against the West Buxton strain (Al), EEV (A3) and AS (A6). All of these serotypes have now been tentatively designated Al to AlO belonging to serogroup A of the aquatic birnaviruses (Christie, Ness & Djupvik 1990).…”

“…With one monoclonal antibody known to be VP3-specific, Wolski, Roberson & Hetrick (1986) have shown that prototype strains within serotype A2 share a common antigenic determinant. Caswell-Reno et al (1989) have shown a common antigenic determinant in strains isolated in different species and geographical areas within serotype Al. Similarly, Christie et al (1990) have shown a common epitope in IPNV isolates within serotype AlO.…”
