Use of the Ussing chamber technique to study nutrient transport by epithelial tissues

Abstract: The Ussing chamber provides a physiologically relevant system for measuring the transport of ions, nutrients, and drugs across various epithelial tissues. This article outlines the design, structure, principle, and operation of the Ussing chamber, its application in the field of gastrointestinal barrier function and nutrient transport research, as well as its advantages and limitations. This review serves as a practical guide for investigators who are new to the Ussing chamber and should help researchers bette… Show more

“…Powerful methodologies include measurements of mRNA and protein (total and phosphorylated) abundances in cells and tissues using quantitative real-time PCR and western blotting techniques. Additionally, as for other nutrients (244,245), transport of 3 H 2 O and ions by cells or tissues in the Ussing chamber system provides a functional indication of AQP abundance and/or activity (Figure 4). Future investigations of AQP in the mammalian female reproductive system require specific AQP blockers/inhibitors or animal models with defined phenotypes for AQP deficiencies.…”

“…The in vivo lysine transport was tested via Ussing chamber according to our previous report [20]. Briefly, 10-cm segments were excised from the proximal jejunum and immediately placed in oxygenated HEPES-Tris buffer solution (pH 7.4) at 37 o C. Mucosa sample were made for 4 replicates respectively, aperture area was set for 0.5cm 2 as the exposed tissue surface area, and the fluid volume in each chamber was 5 ml.…”

“…Prostaglandin E2 (PGE2) and interleukin-1beta (IL-1β) are the substances which are elevated in large intestine diseases and participate in the development of pathologies (Kamiyama et al, 2006;Sheng et al, 2006;Al-Sadi et al, 2012;Carmi et al, 2013;Voronov et al, 2013;Safdari et al, 2016;Zielinska et al, 2018). However, their effect on the barrier properties of different parts of the colon has not been studied yet.…”
