Utilizing Quantitative Proteomics to Identify Species-Specific Protein Therapeutic Targets for the Treatment of Leishmaniasis

Abstract: Leishmaniasis is a tropical disease caused by Leishmania parasites, which are transmitted through the bites of infected sandflies. We focused on the emergence of leishmaniasis in Thailand caused by a species ( Leishmania orientalis ). Treatment by chemotherapy is not effective against L. orientalis . Hence, we intended to solve this issue using a proteomics approach to investigate protein profiles and in silico analysis… Show more

“…These pathways may be related to parasite growth and development and contribute to their response to stress [ 40 42 ]. KEGG pathway analysis is generally used to capture the systematic properties of the inner activities of cells as reference information for the integration and interpretation of high-throughput proteomics datasets [ 43 , 44 ]. Therefore, we performed KEGG pathway enrichment analysis with the aim of discovering important molecules or pathways that play key roles during S. mansoni growth and development.…”

“…During the proteome analysis, 6099 proteins were identified, of which 1065 were used for analysis. In this study, 16 proteins were identified as therapeutic targets for all species, and an in silico analysis of protein antigenicity showed that eight proteins (E8NH10, A4IAU0, A0A640KJ19, A0A451EJM4, ndpk, A0A3S7WPD8, A4HEA3, Q4Q8G4) genes hydrophferently conveyed Pjorlar amastiogote have the potential for the development of antigenic molecules for the treatment of diseases caused by any Leishmania species [ 48 ].…”

“…The alga cells were prepared for proteomics using a previously published protocol with minor modifications [16,17]. Briefly, the cells were lysed on ice using a probe tip sonication at a frequency of 20 kHz and 80% amplitude for 2 s on and 3 s off for a total of 15 s in 200 l lysis buffer (0.2% TritonX-100, 10 mM DTT, 5 mM sodium chloride, 10 mM HEPES-KOH, pH 8.0) with protease inhibitor cocktail.…”
