Vacuolar status and water relations in embryonic axes of recalcitrant Aesculus hippocastanum seeds during stratification and early germination

Abstract: Cells of embryonic axes in recalcitrant horse chestnut seeds are associated with a high water content and functionally preserved vacuoles with active aquaporins and invertase. These vacuoles function to the support rapid cell elongation of the hypocotyl that results in radicle emergence and thus the start of visible germination.

“…Also in the forest species Aesculus hippocastanum aquaporin gene expression is low during the beginning of imbibition and only at radicle emergence it is strongly activated. The authors suggest the presence of aquaporins in the seeds during imbibition phase, but in a closed state (Obroucheva et al 2012;Obroucheva 2013).…”

“…Although the use of mercury can not by itself provide definitive evidence for aquaporins function, it allows us to indicate possible activity and function of these proteins (Willigen et al 2006) and its use is still found in current works (Obroucheva et al 2012;Liu et al 2014). Novikova et al (2014) say that in growing organs, and in the absence of suitable genetically altered materials, studies performed with mercury or other pharmacological blockers remain the only accessible technique for probing the functioning of aquaporins.…”

“…For the desiccationtolerant organisms, drying is often a strategy to avoid rapid deterioration of living tissues and this implies several advantages. In the dry state, orthodox seeds, for example, can be distributed widely (Alpert 2005), since they can be stored for long periods, whereas recalcitrant seeds developed mechanisms for a quick germination (Barbedo & Marcos Filho 1998, Obroucheva et al 2012. In spite of not being immortal (Berjak 2006), seeds of some species can be stored for as long as thousands of years, such as Nelumbo nucifera Gaertn.…”
