Variable Sulfation of Dietary Polyphenols by Recombinant Human Sulfotransferase (SULT) 1A1 Genetic Variants and SULT1E1

Abstract: ABSTRACT:Human cytosolic sulfotransferases (SULTs) catalyze the sulfate conjugation of several important endo-and xenobiotics. Among the superfamily of SULT enzymes, SULT1A1 catalyzes the sulfation of small planar phenolic compounds, whereas SULT1E1 has a major role in estrogen conjugation. The human SULT1A1 gene has common single nucleotide polymorphisms that define three allozymes, SULT1A1*1, *2, and *3. The enzyme kinetics of SULT1A1 allozymes and SULT1E1 were characterized for the polyphenolic substrates a… Show more

“…SULT Isoform (Table 3). SULT1A1, SULT1A3, and SULT1E1 have been reported to sulfonate other flavonoids as well (Otake et al, 2002;Nakano et al, 2004;Ung and Nagar, 2007). In the present study, SULT1A1, SULT1A2, and SULT1E1 solely catalyzed the formation of hesperetin 3Ј-O-sulfate, whereas SULT1C4 and SULT2A1 solely catalyzed the formation of hesperetin 7-O-sulfate (Fig.…”

“…Sulfation reactions were conducted using HLC. In brief, by use of a modified assay (Ung and Nagar, 2007), a total incubation volume of 500 l contained 1 mg/ml HLC, 1 mM MCP, and 4 mM 3Ј-phosphoadenosine-5Ј-phosphosulfate in 10 mM potassium phosphate (pH 6.5). Incubations were performed for 3 h at 37°C and then terminated with ice-cold acetonitrile (200 l).…”

“…RES is extensively metabolized into its sulfated and glucuronidated metabolites, with trans-resveratrol-3-sulfate (R3S) and trans-resveratrol-3-glucuronide (R3G) as major metabolites (Juan et al, 2010;Iwuchukwu et al, 2012;Sharan et al, 2012). RES is sulfated mainly by SULT1A1 with minor contribution from SULT1E1 (Ung and Nagar, 2007). RES is glucuronidated at its 3-OH position via UGT1A1, UGT1A6, UGT1A7, UGT1A8, UGT1A9, UGT1A10, and UGT2B7 (Brill et al, 2006).…”
