Vascular composition, apoptosis, and expression of angiogenic factors in the corpus luteum during prostaglandin F2α-induced regression in sheep

Abstract: Corpora lutea and blood samples were collected from superovulated ewes 0, 4, 8, 12 and 24 h after prostaglandin F 2a (PGF) analog injection on day 10 of the estrous cycle. Changes in vascular cell and fibroblast composition, apoptosis and mRNA expression for several angiogenic factors in the corpus luteum (CL) were determined. While peripheral progesterone concentration decreased at 24 h after PGF injection, CL weight did not change. The area of positive BS-1 lectin staining (endothelial cell marker), smooth m… Show more

“…6). Apoptotic cells were also detected in ovine luteal tissues serving as a control (data not shown; see Vonnahme et al 2006, Grazul-Bilska et al 2008. Appearance of apoptotic cells in fetal ovaries was not affected by nutritional treatment.…”

“…However, while the mRNA expression for VEGFR1 is maintained, expression of VEGFR2 declines on day 18 of the cycle. Similarly, Vonnahme et al (2006) show VEGFR1 mRNA expression unchanged 12 h after PGF2α injection, whereas VEGFR2 was reduced 4 h after treatment.…”

“…Nonetheless, our results are in physiological agreement with the events that occur at luteolysis. In the ovine CL, PGF2α produced an increase in total VEGF mRNA expression 4 and 8 h after its injection, however, 24 h after PGF2α injection the expression of VEGF was below that of CLs collected before treatment (Vonnahme et al, 2006). PGF2α is released by the endometrium from days 15 to 19 of the bovine oestrous cycle (Ginther et al, 2010).…”

“…2). In other species, injection of PGF2α consistently stimulated eNOS mRNA expression and/or NOS activity within the CL of sheep [42], rat [23], and rabbit [22]. Moreover, PGF2α upregulates NOS activity in mature CL (Day 9) but not in immature CL (Day 4) of pseudopregnant rabbits [35].…”
