Vegf-B

Abstract: Despite its early discovery and high sequence homology to the other VEGF family members, the biological function of VEGF-B remained debatable for a long time, and VEGF-B has received little attention from the field thus far. Recently, we and others have found that (1) VEGF-B is a potent survival factor for different types of cells by inhibiting apoptosis via suppressing the expression of BH3-only protein and other apoptotic/cell deathrelated genes. (2) VEGF-B has a negligible role in inducing blood vessel grow… Show more

“…Moreover it is a survival factor for endothelial cells, pericytes, and smooth muscle cells [201, 218, 219]. Administration of small quantities of VEGF-B causes neuroprotection without causing adverse effects or vascular blood-brain barrier leakiness [220].…”

“…Though VEGF-B is not essential for angiogenesis in most tissues and settings, it has been demonstrated to be a crucial survival factor for blood vessels [88, 89]. Due to the fact that VEGF-B shows only minor angiogenic activity, VEGF-B was shown to have a better safety profile as a neuroprotective survival molecule than VEGF-A and might therefore be a good therapeutic target for neurodegenerative diseases [90].…”

“…Blockade of VEGFR-1 eliminates the inhibitory effect of VEGF-B on the expression of anti-apoptotic genes in RGCs, strongly suggesting that VEGFR-1 mediates the neuroprotective properties of VEGF-B in the retina. In contradiction to VEGF-A, VEGF-B lacks a general angiogenic activity (except in the heart), which makes it an ideal candidate for neuroprotective therapies without affecting the vascular system [89]. …”
