Show more

“…Aptamers are either DNA or RNA single strands that can fold into certain active conformations and bind to many target structures with usually high affinities and good selectivities and can hence be used for example as inhibitors. [23,24] Therefore we got interested in preparing caged aptamers and teamed up with the group of Günter Mayer at the University of Bonn who had been studying aptamers for quite some time. One extensively studied aptamer is the aptamer HD1 (Fig.…”

“…By taking advantage of the fact that formation of the RNA-ligand complex increases the stability of an RNA structure compared to that of the RNA by itself, several gene regulation systems were engineered in which Shine-Dalgarno (SD) and initiation codon sequences were selectively sequestered depending on the presence of an exogenously added ligand, which altered the structure of the mRNA upon binding. [4] Such results strongly suggested that the RNA World could have benefited from feedback regulatory mechanisms to regulate gene expression.…”

“…However, it was repeatedly shown by using SELEX (Systematic Evolution of Ligands by EXponential enrichment) that RNA can be a "good contortionist" at building complex architectures. [3] A large number of artificial RNA aptamers have been isolated that respond to various small molecules, such as theophylline, adenosine triphosphate, amino acids and organic dyes (recently reviewed [4] ). By taking advantage of the fact that formation of the RNA-ligand complex increases the stability of an RNA structure compared to that of the RNA by itself, several gene regulation systems were engineered in which Shine-Dalgarno (SD) and initiation codon sequences were selectively sequestered depending on the presence of an exogenously added ligand, which altered the structure of the mRNA upon binding.…”

“…9,10 DNAzymes can be artificially identified from a randomsequence DNA library by in vitro selection [11][12][13][14] and have been increasingly applied as molecular tools for various applications. [15][16][17][18] One powerful type of DNAzyme is an RNAcleaving fluorogenic DNAzyme (RFD).…”
