Virus preparations from the mixed-infected P70 Pinot Noir accession exhibit GLRaV-1/GVA ‘end-to-end’ particles

Abstract: P70 is a Pinot Noir grapevine accession that displays strong leafroll disease symptoms. A high-throughput sequencing (HTS)-based analysis established that P70 was mixed-infected by two variants of grapevine leafroll-associated virus 1 (GLRaV-1, genus Ampelovirus) and one of grapevine virus A (GVA, genus Vitivirus) as well as by two viroids (hop stunt viroid [HSVd] and grapevine yellow speckle viroid 1 [GYSVd1]) and four variants of grapevine rupestris stem pitting-associated virus (GRSPaV). Immunogold labellin… Show more

“…The clearly visible phenotypic characteristics of infection were a darker area around the leaf veins and downward-twisted leaf edges (Martelli 1993, Andret-Link et al 2004, Meng et al 2017. Both symptoms are characteristic of red varieties infected with the most widespread and damaging viruses (Alliaume et al 2018, Sabella et al 2018 of the leafroll complex (Meng et al 2017). In the advanced phase of infection, the viruses of the leafroll complex cause anthocyanin accumulation.…”

“…Virus source grapevines were rooted cuttings from the following accessions of our reference collection of grapevine viruses [ 18 ]: P70 [ 19 , 20 ], Y199, Y217, Y252, Y258, Y318, and Cab119-66. Their virus content was checked by ELISA (see Section 2.5 below) and RT-PCR, as previously detailed in [ 21 ].…”

“…ficus [9], Pseudococcus longispinus (Targioni Tozzetti) [10] and Parthenolecanium corni (Bouché) [11]. As these vitiviruses are frequently transmitted along with GLRaV 1 or 3, the hypothesis that ampeloviruses may assist vitiviruses during transmission has been raised [7,[11][12][13][14]. A semi-persistent transmission mode is believed to apply to soft scales, as already shown for mealybugs (e.g., [7,10,[15][16][17][18][19][20]).…”
