Vitamin B12, also known as the anti-pernicious anemia factor, is an essential micronutrient totally dependent on dietary sources that is commonly integrated with food supplements. Four vitamin B12 forms—cyanocobalamin, hydroxocobalamin, 5′-deoxyadenosylcobalamin, and methylcobalamin—are currently used for supplementation and, here, we provide an overview of their biochemical role, bioavailability, and efficacy in different dosage forms. Since the effective quantity of vitamin B12 depends on the stability of the different forms, we further provide a review of their main reactivity and stability under exposure to various environmental factors (e.g., temperature, pH, light) and the presence of some typical interacting compounds (oxidants, reductants, and other water-soluble vitamins). Further, we explore how the manufacturing process and storage affect B12 stability in foods, food supplements, and medicines and provide a summary of the data published to date on the content-related quality of vitamin B12 products on the market. We also provide an overview of the approaches toward their stabilization, including minimization of the destabilizing factors, addition of proper stabilizers, or application of some (innovative) technological processes that could be implemented and contribute to the production of high-quality vitamin B12 products.

Archiv der Pharmazie 2~Alkyl-4-acylpyridinthiosemicarbazone (VI)0,060 Mol 2-Alkyl-4-acylpyridin und 0,066 Mol Thiosemicarbazid wurden in 150 ml Wasser suspendiert und 16 Std. zum Sieden erhitzt. Nach Abktihlen wurde abgesaugt, der Filterriichtand zweimal mit je 60 ml heil3em Wasser gewaschen und a m n-Propylalkohol umkristallisiert. Die 2-Alkyl-4-acylpyridinthiosemicarbazone wurden in Form von achwach gelblich gefirbten Kristallen erhalten. Die Darstellung der hellgelb gefiirbten 2-Alkyl-4-acylpyridinthiosemicarbazon-hydrochloride erfolgte durch Behandeln mit methanol. Salzsiiure. (Eingegangeu a m 21. Juli 1964) Versuohe ergaben, daB die Pyrimidin-Komponente des Aneurins, beaonders in Verbindung mit einem Phosphatpuffer, ein auagezeichneter Stabilisator fiir Cyanocobalamin in Cegenwart von Aneurin und Nikotinsgureamid ist. Es wird damit eine Haltbarkeit sowohl der Losungen mit 0,l und 0,05y0 Cyanocobalamin als auch mit nur O,Olyo Cyanocobalamin nach Gwochiger Lagerung bei 37' erreicht, die einer Haltbarkeit von etwa 2 Jahren bei Raumtemperatur von 20 bis 24' entspricht. Die Haltbarkeit erstreckt sich nicht nur auf den Cyanocobalamin-Gehalt, sondern auch auf die Farbe der Losung. Bei Lagerung im KWchrank von 1 bis 5" kann mit einer noch liingeren Haltbarkeit gerechnet werden. Mit der Pyrimidin-Komponente und mit Phosphatpuffer wird somit die stabilisierende Wirkung des Kaliumrhodanids, beaonders in der Lasung mit nur O,Olyo Cyanocobalamin, weit iibertroffen. Von den vielseitigen Vitamin-Kombinationen fiir parenterale Losungen hat in den letzten Jahren die zwischen Aneurin und Cyanocobalamin allein oder in Gegenwart anderer Arzneistoffe die Aufmerksamkeit in betrachtlichem MaBe auf aich gezogen, wed mehr oder weniger grode Cyanocobalamin-Verluste damit verbunden sind. Die Arbeiten zur Stabilitiit von Cyanocobalamin in Gegenwart anderer Vitamine zeigen in vieler Hinaicht ein widerspruchsvolles Bild. Nach Macek und FeZkr1) aus l) T. J . Mecelc und B. A.
