Voluntary Sleep Loss in Rats

Abstract: We conclude that ICSS and NCS can be used to sleep deprive rats. Changes in rebound NREM sleep EEG SWA occurring after ICSS, NCS, and GH-SDEP suggest that nonspecific effects of the SDEP procedure differentially affect recovery sleep phenotypes.

“…Briefly, when a mouse became motionless a soft artist's paintbrush was used to tap the cage or disturb the bedding around the animal to keep it awake and, when necessary, the back of the mouse was gently stroked with the brush. Sleep-rebound durations after gentle handling of SD are similar to those after voluntary SD induced by protracted intracranial selfstimulation in animals (45). Body temperature and locomotor activity recordings continued for 48 h of recovery.…”

“…Interestingly, delta power during NREM sleep (but not during time awake) was elevated following administration of VU0424238 but not by M-5MPEP (Supplementary Figures 2, 3, 5). NREM sleep duration during the subsequent 12 h dark cycle was increased following VU0424238 but not M-5MPEP, likely reflecting the NREM sleep rebound following acute wake-promoting effects (Franken et al, 1991;Oonk et al, 2016;Dispersyn et al, 2017). While insomnia has been reported in a majority of patients with MDD, hypersomnia is also reported in a smaller subset of patients (Armitage, 2007;Steiger and Pawlowski, 2019).…”

“…Animals were sleep deprived for 12h (ZT0-12) by gentle handling as previously published [ 22 ]. Animals were stimulated by gently touching the tail, body, or nose of the animal with a wooden tongue depressor or small paintbrush when the animal appeared to fall asleep or when EEG/EMG indicated a transition to a sleep state.…”
