Wavelength dependence of cell cloning efficiency after optical trapping

Abstract: A study on clonal growth in Chinese hamster ovary (CHO) cells was conducted after exposure to optical trapping wavelengths using Nd:YAG (1064 nm) and tunable titanium-sapphire (700-990 nm) laser microbeam optical traps. The nuclei of cells were exposed to optical trapping forces at various wavelengths, power densities, and durations of exposure. Clonal growth generally decreased as the power density and the duration of laser exposure increased. A wavelength dependence of clonal growth was observed, with maximu… Show more

“…According to previous studies, laser wavelength is a critical parameter determining photochemical damage at the cellular level. 43 Laser-induced damage can be estimated by the inverse of the relative reduction in cloning efficiency. Cell damage induced by 808 nm is reduced by about 30% with respect to that caused by 790 nm radiation.…”

“…Using light to manipulate objects with high precision and finesse can revolutionize multidisciplinary research fields. This technique made it possible to reach lower temperatures in atomic cooling [117][118][119], to achieve higher resolution in light scattering [120], to manipulate living biological cells at will without causing damage [121][122][123], and to fabricate complex nanoscale structures [124].…”

“…For instance, photodamage is the lowest at the wavelength of 830 nm for E.coli 136 but at 970 nm for Chinese hamster ovary cells. 137 Therefore, it is necessary to study the optical damage to the biological system on a case-by-case basis. 133 Another drawback of laser trapping is the occurrence of local heating at the focus volume.…”
