Whole-brain R1 predicts manganese exposure and biological effects in welders

Abstract: Manganese (Mn) is a neurotoxicant that, due to its paramagnetic property, also functions as a magnetic resonance imaging (MRI) T1 contrast agent. Previous studies in Mn toxicity have shown that Mn accumulates in the brain, which may lead to parkinsonian symptoms. In this article, we trained support vector machines (SVM) using whole-brain R1 (R1 = 1/T1) maps from 57 welders and 32 controls to classify subjects based on their air Mn concentration ([Mn] Air ), Mn brain accumulation (ExMn Brain ), gross motor dysf… Show more

“…However, another study did not observe any association between thalamic and striatal GABA levels and blood Mn or airborne metal exposure levels in welders [ 186 ]. This inconsistency may be explained by non-linear association between Mn exposure and brain GABA alterations, with the latter being undetectable at lower doses and periods of exposure [ 187 ].…”

“…Currently, the signal intensity on a T1-weighted MRI for the globus pallidus, caudate and putamen and pallidal index is the most reliable MRI-related biomarker for the diagnosis of manganese toxicity [24]. The T1-weighted basal ganglia signals and pallidal index are associated with occupational Mn exposure and the severity of Parkinsonism in steelworkers, miners and welders [25][26][27]. However, these MRI abnormalities are not directly related to the symptoms of manganism, as they can also occur in asymptomatic welders [28].…”
