Within-Subject Reliability and between-Subject Variability of Oxidative Stress Markers in Saliva of Healthy Subjects: A Longitudinal Pilot Study

Abstract: The present study evaluated diurnal variations and day-to-day fluctuations of salivary oxidative stress (OS) markers in healthy adult individuals. Whole unstimulated saliva was collected at 2 time intervals over 3 consecutive days. Glutathione peroxidase (GPX), superoxide dismutase (SOD), total antioxidant capacity (TAC), and uric acid (UA) were analyzed using spectrophotometric methods, while 8-hydroxydeoxyguanosine (8-OHdG) and malondialdehyde (MDA) were determined using immunoassays. No significant differen… Show more

“…Urinary biomarkers of oxidative stress were measured in samples from 4 study visits across pregnancy indicating variable unadjusted 8-oxodG concentrations with ICC = 0.32 38 . Saliva of healthy adults was collected over 3 consecutive days to assess unadjusted 8-oxodG diurnal variations and intra-individual CVs of 40% samples was higher than 35%, which was calculated by the following equation, (between-day SD/between-day mean) × 100, indicating high intra-individual variability 39 . SG and creatinine were proved to be temporal consistent in urina sanguinis collected from 243 subjects over 5 consecutive weekdays 40 .…”

“…The methods of sample collection and analysis were described in detail and validated by our group in a prior study ( Alajbeg et al, 2017 ). The subjects were instructed to fast before saliva collection in the morning and to not eat or drink anything but water at least 2 h before sampling in the afternoon.…”
