Wnt-11 signalling controls ventricular myocardium development by patterning N-cadherin and β-catenin expression

Abstract: We conclude that Wnt-11 signalling serves as a critical cell adhesion cue for the organization of the cardiomyocytes in the developing ventricular wall, which is essential for the establishment of a functional heart.

“…Of interest, Wnt11R is a true ortholog of other vertebrate Wnt11 genes . Genetic analyses have demonstrated a role for Wnt11 in the ureteric branching of the mouse kidney (Majumdar et al, 2003) and in heart morphogenesis (Nagy et al, 2010), but its role in NC development has not been examined. Consistent with the role in NC specification, Xenopus Wnt11R is expressed in the neural plate, adjacent to the NC territory (Matthews et al, 2008a).…”

“…74,75 This putative functional redundancy might explain why a loss of Wnt11 results in an early cardiac phenotype in Xenopus 49,51 but not in the mouse with later phenotypes. 76,77 Slightly later, however, it might be Wnt2b that regulates proliferation of the common cardiac progenitor through a Wnt/␤-catenin pathway. 78 enter the heart tube either from the anterior site giving rise to the right ventricle and the OFT or they can enter from the posterior site contributing to the atria and the inflow tract region.…”

“…A detailed analysis indicated defects in N-cadherin/␤-catenin mediated cell adhesion as well. 76 Similarly, Wnt5a promotes N-cadherin-mediated cell adhesion in cardiac myocytes. 94 Besides N-cadherin, the cell adhesion molecule DM-GRASP also has been shown to be regulated by noncanonical Wnt signaling in Xenopus.…”

“…The role of Wnt11 in cell motility was reported related to the directional movements of the cells in the assembling ventricular wall by modulating cell adhesion in connection with changes in the cytoskeleton (Nagy et al, 2010). A common theme is that Wnt11 acts locally to control the turnover of proteins involved in cell-cell and cell-substrate adhesion and thereby facilitate coordinated cell migration (Katoh, 2009;Uysal and Kypta, 2011).…”
