Xanthohumol‐supplemented beer modulates angiogenesis and inflammation in a skin wound healing model. Involvement of local adipocytes

Abstract: Angiogenesis and inflammation are two intermingled processes that play a role in wound healing. Nevertheless, whenever exacerbated, these processes result in nonhealing wounds. Xanthohumol (XN), a beer-derived polyphenol, inhibits these processes in many physiopathological situations. This study aimed at examining whether XN ingestion affects wound healing. Wistar rats drinking water, 5% ethanol, stout beer (SB) or stout beer supplemented with 10 mg/L XN (Suppl SB) for 4 weeks, were subjected to a 1.5 cm full … Show more

“…Chalcone xanthohumol (XN) is the major prenylated flavonoid found in natural plant, Humulus lupulus [1]. It was first isolated by Power et al [2] and its molecular structure consists of two aromatic rings (A and B) substituted with hydroxyl and methoxyl groups, one ␣, ␤ unsaturated double bond, and the prenyl unit [3,4] (Scheme 1).…”

“…Briefly, starting from acetylation of commercially available naringenin, the 7,4’‐diacethyl‐naringenin derivative was O ‐prenylated by the Mitsunobu reaction and following the tandem Claisen‐Cope rearrangement of 5‐prenyl‐7,4‐diacetylnaringenin with Eu(fod) 3 , 8PN was obtained in 32% yield. XN and 8PN were administered in a dose of 10 mg/L previously studied by our group, equivalent to 1 mg/kg/day, which is in line with the doses reported previously without displaying hepatotoxic effects . Polyphenols consumption was administered throughout the 20 weeks of experiment, the duration necessary to establish hyperglycemic conditions.…”

“…[14][15][16] In a rat skin wound healing model, systemic administration of stout beer or stout beer supplemented with 10 mg/L xanthohumol for 4 weeks was found to significantly reduce inflammation, oxidative stress, and angiogenesis, ameliorating the wound healing process, as compared with the control group drinking only water. 17 Xanthohumol was also shown to significantly inhibit elastase and matrix metalloproteinase (MMP)-9 activities (less effective on MMP-1 and MMP-2) and dramatically increase the expression of collagens type I/III/V and elastin, fibrillin-1, and fibrillin-2 in dermal fibroblasts, to similar degree of treatment with ascorbic acid. 18…”
