Introducción. Es bien sabido que, entre los caracteres transmitidos por la línea paterna, el apellido se ha configurado en diferentes culturas como un carácter semejante a un alelo genético neutral asociado al cromosoma Y.Objetivo. En este estudio se determinaron las frecuencias en la población de 17 STR del cromosoma Y en 308 individuos provenientes de las poblaciones de los departamentos del Valle del Cauca, Cauca y Nariño. Además, se propuso definir la correlación de los haplotipos obtenidos en cada individuo con su apellido paterno, para comparar estos dos códigos de identidad. Materiales y métodos. Se extrajo el ADN de cada individuo a partir de sangre periférica y se utilizó el estuche comercial AmpFLSTR ® Yfiler™ (Applied Biosystems) para tipificarlo. Los resultados de los haplotipos moleculares se compararon con los apellidos reportados por cada individuo y se asociaron apellidos amerindios y europeos con haplotipos que incluyeran o no el marcador DYS19/13, característico de la población amerindia. Resultados. Se reportan las frecuencias alélicas de cada uno de los 17 marcadores del cromosoma Y analizados en esta región de Colombia, así como la diversidad génica y haplotípica hallada en los tres departamentos. Al comparar los resultados obtenidos a nivel molecular con los apellidos de origen europeo o amerindio reportados por cada uno de los individuos, se encontró cerca de 40% de inconsistencia de linaje. Conclusiones. La utilización del apellido como marcador de población debe hacerse con cautela, por cuanto las genealogías fundamentadas en éstos pueden no corresponder al origen biológico de sus portadores.Palabras clave: genotipo, nombres, cromosoma Y, indios sudamericanos, Colombia. Correlation analysis of surnames and Y-chromosome genetic heritage in 3 provinces of southwestern ColombiaIntroduction. In Colombia, surnames are characters usually passed to the children by the father, and they have been compared to neutral alleles associated with the Y-chromosome. Objective. Population frequencies were determined for 17 short tandem repeats (STR) DNA markers on the Y-chromosome to compare the two identity codes and define the correlation between haplotypes and surnames in each individual. Materials and methods. DNA was extracted from blood samples from 308 male individuals in provinces of Valle del Cauca, Cauca and Nariño, all in southwestern Colombia. In the Pacific and Caribbean coast different degrees of admixture between African descent and Caucasian-Mestizos is present. On the other side, Amerindian admixture is stronger in the southwest and decreases towards the north section of the Andes were the Caucasian component is stronger.In the present study, we have analyzed the Y-chromosome haplotype data from 271 unrelated individuals, 134 of African descent and 137 Caucasian-Mestizos of Colombia in order to determine the Y chromosome haplotype origin of the two population groups tested. Additional analysis were carried out in order to determine the degree of genetic admixture between these groups and Amerindian populations of Colombia studied by us (unpublished results) and to correlate our findings with previously defined populations of Europe and North America. Material and methodsOne hundred and thirty four (134) unrelated individuals of African descent were collected between 1994-1995, after proper informed consent was obtained, in four different towns (Quibdo n=40, the state capital; Condoto n=34; Istmina n=30; and Tado n=30) of the Choco department in the Pacific region of Colombia. This was part of a research project at the Universidad Nacional de Colombia with the objective to characterize genetically different ethnic groups. In addition, a total of 177 Caucasian-Mestizo individuals from the East-central Andean Region of Colombia (Cundinamarca, n=40, Boyacá ,n=40, and Santander, n=40, departments, as well as Bogotá, n=50, the capital city) divided as 45 alleged father/son pairs with known exclusion of paternity, 40 alleged father/son pairs with proven paternity (minimum of 12 STRs and W>99.99%), and 7 unrelated individuals were analyzed, for a total of 137 unrelated Caucasian-Mestizo individuals were provided by Servicios Médicos Yunis Turbay y Cía., after proper informed consent was obtained in order to establish Y STR haplotype frequencies, that could be used for forensic estimations by many laboratories in Colombia and abroad. Mestizos currently represent most of the Mexican population (>90%); they are defined as individuals born in the country having a Spanish-derived last name, with family antecedents of Mexican ancestors back at least to the third generation. Mestizos are result of 500 years of admixture mainly among Spaniards, Amerindians, and African slaves. Consequently, a complex genetic pattern has been generated throughout the country that has been scarcely studied from the paternal point of view. This fact is important, taking into account that gene flow toward the New World comprised largely males. We analyzed the population structure and paternal admixture of present-day Mexican-Mestizo populations based on Y-STRs. We genotyped at least 12 Y-STRs in DNA samples of 986 males from five states: Aguascalientes (n = 293); Jalisco (n = 185); Guanajuato (n = 168); Chiapas (n = 170); and Yucatán (n = 170). AmpFlSTR Y-filer and Powerplex-Y(R) kits were used. Inclusion of North and Central Y-STR databases in the analyses allowed obtaining a Y-STR variability landscape from Mexico. Results confirmed the population differentiation gradient previously noted in Mestizos with SNPs and autosomal STRs throughout the Mexican territory: European ancestry increments to the Northwest and, correspondingly, Amerindian ancestry increments to the Center and Southeast. In addition, SAMOVA test and Autocorrelation Index for DNA Analysis autocorrelogram plot suggested preferential gene flow of males with neighboring populations in agreement with the isolation-by-distance model. Results are important for disease-risk studies (principally male-related) and for human identification purposes, because Y-STR databases are not available on the majority of Mexican-Mestizo populations.
