Background: The conjunctiva of the eye is a mucosal surface that is colonized by various bacteria. Following surgeries, trauma, or other underlying conditions the normal flora of these bacteria may cause some eye infections such as conjunctival, corneal infections, or endophthalmitis. Objectives: This study aimed to investigate the bacterial population and antibiotic resistance of the conjunctiva in healthy horses. Study design: Random sampling of ocular samples from 20 horse during the winter (2019). Methods: Swab samples were collected from the eyes of 20 healthy horses of different breeds, ages, and sexes in Tehran province, Iran. The swabs were cultured on blood agar and MacConkey agar and incubated at 37°C for 24-48 hours. The isolated bacteria were characterized using cellular morphology, gram-staining, and biochemical tests. Sensitivity and resistance to 10 different antibiotics were measured using an antibiogram test. Results: Results showed that the bacterial population consisted of various species, with Bacillus cereus being the most frequent. There were no significant differences in the frequency of isolated bacteria between sexes and age groups. There were no significant differences in the sensitivity of bacterial flora of the eyes to Colistin, ceftiofur, florfenicol, amoxicillin, and ampicillin between neither different age groups nor different sexes of the horses (P>0.05). However, sensitivity to enrofloxacin and ciprofloxacin was significantly correlated with the age groups of the horses (P>0.05). Significant difference in sensitivity to trimethoprim-sulfamethoxazole between various sexes of horses was observed. Main limitations: The non-uniformity of the follow-up examination reliance owners or caregiver of the horse for follow-up information and Lack of adequate financial resources to study more horses. Conclusions: The study suggests that the frequency of bacterial flora in horses’ eyes is influenced by housing and management conditions rather than age and sex. In equine ophthalmology, ulcerative keratitis is among the most common conditions and, in general, arises as a consequence of some trauma suffered. Secondarily, subsequent contamination by pathogenic or resident bacteria of the horse’s ocular microbiota may have undesirable consequences. Under physiological conditions, the normal microbiota coexists with the immune status of the host, serving as a barrier, ensuring the health of the ocular surface, and inhibiting the proliferation of pathogens. However, in the imbalance of immune barriers, the normal microbiota can become pathogenic and lead to infection, acting as an opportunistic agent. The present study aims to demonstrate the antimicrobial effect of platelet-rich plasma (PRP), its time of action, and its correlation with the concentration of its same components in vitro on Staphylococcus sciuri, a bacterium with high prevalence in the normal ocular microbiota of horses in the municipality of Minas Gerais. For the preparation of the PRP, eight adult Quarter Horse (QH) horses were used. The individual PRP was prepared by the double centrifugation protocol, and then, the PRPs were added to a pool, followed by testing their interaction in culture with Brain Heart Infusion (BHI) broth at different dilutions against five strains collected from different animals. After 3, 6, 12, and 18 hours, the colony formation units (CFU) count on a 5% horse blood agar plate was evaluated for each time point. Our study showed that Staphylococcus sciuri, the resident microorganism of the ocular conjunctival microbiota of horses, is more susceptible when compared to the standard strain “American Type Culture Collection” (ATCC-29213) Staphylococcus aureus, a pathogenic microorganism, which was used for the validation of our study. The antibacterial effect shown in this study was bacteriostatic for up to 6 hours. The most concentrated PRP dilutions, 1 : 1 and 1 : 2, were also most effective, suggesting that the antibacterial effect is volume dependent.

Oküler flora atın yaşadığı ortam, iklim, coğrafya vb birçok etkenden etkilenebilir. Bu çalışmanın amacını XXX Hipodromunda yarış koşan ve hipodroma yakın çiftliklerde bulunan sağlıklı atlarda bakteriyel ve fungal oküler florayı belirlemek ve belirlenen bakteri ve mantar türlerini tanımlamak amaçlandı. Çalışma materyalini değişik yaş ve cinsiyette XXX Hipodromunda bulunan ve yarış koşan 28 ve hipodrom yakınlarındaki çiftliklerde yarış koşmayan 28 safkan Arap ve İngiliz atına ait 112 sağlıklı göz oluşturdu. Sağlıklı gözlerin medial kantusundan steril swaplarla sürüntü alınarak soğuk zincirde Dicle Üniversitesi Veteriner Fakültesi Mikrobiyoloji Laboratuvarına ulaştırılarak konjunktival florada bulunan bakteri ve fungal etkenlerin izolasyonu ve identifikasyonu yapıldı. Hipodrom grubunda bakteriyolojik üreme %94.64, fungal üreme %28.57, çiftlikte grubunda bakteriyolojik üreme %100, fungal üreme (%14.29) tespit edildi. Değerlendirilen konjunktival svap örneklerinin izole ve identifiye edilen bakterilerin hipodrom grubunda %87.79’unun Gram pozitif %12.21’inin Gram negatif, çiftlik grubunda ise %82.56’sının Gram pozitif, %17.43’ünün de Gram negatif olduğu belirlendi. Her iki grupta da mikrofloranın büyük çoğunluğunu Stafilokok spp. (birbirinden farklı) oluştuğu görüldü. Sonuç olarak, belirli aralıklarla konjunktival floranın belirlenmesi ve olası bir kornea veya göz hasarında izlenecek tedavinin belirlenmesine yardımcı olacağı ve erken müdahale ile kornea hasarına bağlı görme kayıplarının önüne geçilebileceği düşünülmektedir.
