Zerumbone-Induced Antinociception: Involvement of the l-Arginine-Nitric Oxide-cGMP -PKC-K+ATP Channel Pathways

Abstract: This study investigated the antinociceptive effects of zerumbone in chemical behavioural models of nociception in mice. Zerumbone given through intraperitoneal route (i.p.) produced dose-related antinociception when assessed on acetic acid-induced abdominal writhing test in mice. In addition, the i.p. administration of zerumbone exhibited significant inhibition of the neurogenic pain induced by intraplantar (i.pl.) injection of capsaicin and bradykinin. Likewise, zerumbone given by i.p. route reduced the nocic… Show more

“…Zerumbone (1), a bioactive sesquiterpene used in this study was isolated from the rhizomes of Zingiber zerumbet. In our two recent publications, it was shown that zerumbone attenuated pain in animal models of acute pain [20,21]. On another publication, we also demonstrated that zerumbone possesses significant anti-inflammatory properties [22].…”

“…Zerumbone was kept at − 20°C prior to use and was freshly prepared by dissolving with dimethylsulfoxide (DMSO), Tween 20 and normal saline (0.99% NaCl) at a ratio of 5:5:90 (v/v) [21]. The final concentration of DMSO did not exceed 1% and caused no detectable effect 'per se'.…”

“…Then the nociceptive responses against 0.6% acetic acid injection were observed for 30 min, starting from 5 min post injection. The numbers of abdominal writhing were counted as indication of pain behaviour (Abacioglu et al, 2000;Perimal et al, 2011).…”

“…Possible contribution of K + channel in the antinociceptive effect of MEDR was evaluated by established method Perimal et al, 2011). The mice were pretreated with glibenclamide (10 mg/kg), an ATP-sensitive K + channel inhibitor, intraperitoneally 15 min before administration of either vehicle or MEDR.…”

“…(essential oil)에 포함되어 있는 주요 성분인 zerumbone은 혈 액종양을 포함한 암세포 죽음, 염증 및 활성산소 감소 등에 기여 할 수 있음이 보고 되었다 [2][3][4]. 또한 면역세포들의 증식과 세포 주기진행, 사이토카인의 생성·발현에도 효과를 나타낸다고 알 려져 있다 [5].…”
