Zum Nachweis toxinogener Bacillus cereus-Stämme

Abstract

Summary Detection of toxinogenic Bacillus cereus Investigations into optimal culture conditions for Bacillus cereus in order to detect bacterial toxins in a cell culture system showed that Dulbecco's modified Eagle's medium supplemented with 5 % fetal calf serum is the medium best suited for this purpose. The highest toxicity levels were seen when bacteria were cultured at 21 °C. In 42 of 43 Bacillus cereus‐strains isolated from milk and milk products, toxin formation was detected using the MTT‐test. In 11 strains, toxin formation was even shown when bacteria were cultured at 4 °C. When culture supernatants were examined in a commercially available ELISA, all cytotoxic strains were shown to form diarrheal toxin. Zusammenfassung Untersuchungen zur Optimierung der Anzuchtbedingungen für Bacillus cereus zum Nachweis von Toxinen mittels Zellkultursystem zeigten, daß Dulbecco's Modified Eagle's Medium mit Zusatz von 5 % Fetalem Kälberserum sich zu diesem Zweck als am geeignetsten erweist. Die größten Toxizitäten wurden bei einer Anzuchttemperatur von 21 °C erreicht. 42 von 43 aus Milch und Milchprodukten isolierte B. cereus‐Stämme konnten mit Hilfe des MTT‐Testes als Toxinbildner eingestuft werden. Bei 11 Stämmen war eine Toxinbildung auch nach Anzucht bei 4 °C nachweisbar. Die Prüfung der Kulturüberstände in einem kommerziellen ELISA ergab, daß sämtliche im Zelltest als Toxinbildner erkannte Stämme Diarrhoetoxin bilden.
