α-Mangostin reduced the viability of A594 cells in vitro by provoking ROS production through downregulation of NAMPT/NAD

Abstract: α-Mangostin (MAN) is a bioactive compound isolated from the inedible pericarp of a tropical fruit mangosteen (Garcinia mangostana Linn). It exhibits notable therapeutic potentials on lung cancers, but the underlying mechanisms are still largely unknown. This study was designed to further explore the mechanisms involved in cytotoxicity of MAN on A549 cells. Apoptosis and cell cycle distribution were analyzed by flow cytometry methods. The fluorescent probes DCFH-DA and JC-1 were used to assess the intracellular… Show more

“…α-mangostin has anti-cancer and anti-proliferative effects on many types of cancers 21 . Existing evidence indicates that α-mangostin generally induces cell cycle arrest and apoptosis in cancer cells 22 . α-mangostin accelerates the apoptosis of mouse skin tumor cells induced by 9,10-dimethylbenz[a]anthracene (DMBA)/TPA Perish 7 .…”

“…Finally, we used NR, a pyridine-nucleoside form of vitamin B that functions as a NAD precursor [40], to assess whether NR can recover the negative effects arising from NKO. NR has been proven to effectively increase NAD synthesis in various tissues and organs such as the heart, skeletal muscle, adipose tissue, and liver [19,29,41,42]. Our data showed that NR supplementation significantly increased Nampt and NAD levels in HFD-fed mice.…”
