Microflora of the environment plays an important role in the formation of the microflora of the digestive tract of fishes (Strom and Olafsen Microbiology of poecilotherms Elsevier, Amsterdam, pp 181-185 1990; Hansen et al. Appl Environ Microbiol 58:461-470 1992). Compared to water, digestive tract is an ecosystem far richer in nutrients and therefore more favorable for the growth of the majority of bacteria. Definitely, not all bacteria in food which gain entry in the digestive tract of fishes establish themselves there (Yan-Bo Wang et al. Aquaculture 281(1-4): 1-4 2008).Part of them adapts themselves in the digestive tract, whereas the others are digested by the enzymes produced by the host organism. Microorganisms present in the digestive tract feed on the food of the host organism which is digested by the enzymes produced by them and by the latter. 'Chymous' gets formed which decides the abundance and qualitative composition of microorganisms present in digestive tract.
:The work was planned to develop diagnostic assay using hyper immune sera and poultry sera with seroprevalence monitoring in Ranchi (Jharkhand). The best result for dot-ELISA was standardized with 1.2 l of BursaB2K and Gumboro strain, skimmed milk 1.5 per cent+gelatin 0.5 per cent+BSA 1 per cent as blocking solution, sera dilution of 1:10 and 1:500 conjugate dilution. Statistical analyses were showing there is a non-significant difference in results of dot-ELISA and ELISA test to detect specific anti IBDV antibodies. : ELISA and AGID based seroprevalence study of 92 suspected poultry samples in and around Ranchi district of central and western plateau agro-climatic zone in and around Ranchi district, Jharkhand during 2014-15 were 93.48 per cent and 53.26 per cent, respectively. The sensitivity and specificity of AGID was 52.33 per cent and 33.33 per cent, respectively taking ELISA as gold standard test. ELISA and AGID were found significant on standard method of statistical analysis. As per available literature seroprevalence of IB has not been reported by any worker from Ranchi, Jharkhand, however individual cases had been reported. It seems to be the first report from this area which clearly indicates extremely high IB seroprevalence without showing gross pathological lesion.
