Present study focused on evaluation of the effect of national nancomposite product on serum biochemical profile and programmed cell death (apoptosis) by using Annexin V-FITC and PI test of breast carcinoma female rats. Basic nanocurcumin was tested for its protective effect on breast cancer female rats (n= 30) divided in two main groups (NTBR) and (TBR) which in turn classified into (TBR-Control Positive and TBR-basic nano-curcumin protective groups). Basic nano-curcumin was formulated and tested for both IC50 (IC50 = 1.0 µg) and LD5o (35 mg/kg by wt.) by using in vitro and in vivo techniques, respectively. Breast cells apoptosis was determined by Annexin V-FITC, which is a fluorescent probe binds to phosphotidylserine (PS) in a Ca 2+ dependent pathway. Breast Cells were stained with Annexin V and PI. The apoptotic cells [Annexin (+) and PI (-)] were differentiated from necrotic cells [Annexin (-) and PI (+)] using flow cytometry technique. The anti-proliferative effect of basic nano-curcumin appeared much earlier due to its nano form. Also, Liver [ALT (40U/l), AST (85 U/l)] and kidney [BUN (19 mg/dl), Creatinine (0.5 mg/dl)] functions were decreased within normal ranges.
A B S T R A C TThe ameliorative effect and antitumor activity of basic curcumin nanoparticles modified with basic nano black seeds (nigella sativa) and calcium ascorbate were evaluated. eighty female mice were divided into 4 equal groups of 20 mice each. Group 1: non -tumor bearing mice (NTBM). Group 2: tumor bearing mice(TBM). Group 3: NTBM + nanocomposite (4.750gm / kg. b.wt /day ) , orally for 6 weeks . Group 4: TBM + nanocomposite (4.750gm / kg. b.wt/day ), orally for 6 weeks . Blood samples for serum separation and liver tissues specimens were collected from all animal groups three times after 2, 4 and 6 weeks from the onset of treatment. All serum was directly used for determination of ALT and AST activities and urea and creatinine concentrations. Moreover, antioxidants enzymes (catalase, GPX, SOD), GSH and MDA were detected in liver tissue. The obtained results showed significant decrease in catalase, GPX and SOD activities and GSH concentration in liver tissue of tumor bearing mice. Meanwhile, liver MDA, serum transferases(ALT and AST) activities, urea and creatinine concentrations were significantly increased in tumor bearing mice . TBM treated with nanocomposite improve the adverse biochemical changes induced by tumor. So, this nanocomposite has potential benefits in cancer treatment.
Breast cancer is the second leading cause of cancer death in women, There are several ways to treat breast cancer .The effect of Lysine-Cetrimonium gallium Complex with zinc oxide nanopatricle and sodium ascorbate (NNC) on DMBA induced mammary carcinogenesis was investigated in female rats. Oral dose of DMBA induced a significant increase in ALT, AST, MDA and serum NO levels. On other hand marked depletion in CAT, GSH. On other hand oral administration of NNC was able to mitigate mammary carcinogenic damage induced by DMBA as evidence to pronounce curative effect against lipid peroxidation, serum NO level and deviated enzymatic variables and maintained glutathione status, antioxidant enzymes in addition histopathological changes directed toward control. We observed that the administration of (NNC) have potential to exert curative effects against mammary carcinogenesis.
