Endophytic fungi inhabit vegetable tissues or organs, without causing them any harm. Endophytes can co-evolve with plant hosts and possess species-specifi c interactions. They can protect the plant from insect attacks and diseases, and are also able to produce substances of biotechnological interest. In folk medicine, the bark, roots and fruits of Sapindus saponaria is used to produce substances with anxiolytic, astringent, diuretic and expectorant properties, as well as tonics, blood depuratives and cough medicine. This study evaluated the diversity of endophytic fungi present in the leaves of S. saponaria L. and observed the colonization of host plants by endophytes, using light and scanning electron microscopy. We verifi ed that these fungi are found in intercellular and intracellular spaces. The genera of some isolates of S. saponaria were identifi ed mainly by sequencing of ITS region of rDNA and, when possible, also by their microscopic features, as follows: Cochliobolus, Alternaria, Curvularia, Phomopsis, Diaporthe and Phoma. Phylogenetic analysis showed the existence of genetic variability of the genera Phomopsis and Diaporthe and interspecifi c variation among the Curvularia, Alternaria and Phoma, belonging to family Pleosporaceae.
BackgroundChickens are animals that are sensitive to thermal stress, which may decrease their production level in terms that it affects feed intake and thus, decreasing body weight gain. The Heat Shock Factors (HSF) and Heat Shock Proteins (HSP) genes are involved in the key cellular defense mechanisms during exposure in hot environments. Aimed with this study to analyze the expression of HSF1, HSF3, HSP70 and HSP90 genes in two local breeds (Peloco and Caneluda) and a commercial broiler line (Cobb 500®) to verify differences in resistance of these chicken to Heat stress treatment. Chicken were submitted to heat stress under an average temperature of 39°C ± 1.ResultsUnder stress environment, the HSP70 and HSP90 genes were more expressed in backyard chickens than in broiler. There was a difference in HSP70 and HSP90 expression between Caneluda and Cobb and between Peloco and Cobb under stress and comfort environment respectively. HSP70 expression is higher in local breeds during heat stress than in a commercial broiler line. No significant differences were observed in the expression of HSF1 and HSF3 genes between breeds or environments.ConclusionsHSP70 and HSP90 genes are highly expressed, HSF1 and HSF3 genes did not have high expression in all genetic groups. HSP70 and HSP90 are highly expressed in Peloco and Caneluda within heat stress, these breeds proved to be very resistant to high temperature.
Endophyte microorganisms are organisms that live inside plants without causing any apparent damage to their hosts. Since all plants exhibit endophyte microorganisms, it is believed that mutual association is of great importance in nature. Luehea divaricata (Martius & Zuccarini), known popularly in Brazil as açoita-cavalo, is a big-sized tree with a wide distribution in the country that possesses medicinal qualities for: dysentery, leucorrhea, rheumatism, blennorrhoea, tumors, bronchitis, and depuration. This research aims at isolating and molecularly characterizing fungi isolates from L. divaricata by sequence analysis of ITS1-5.8S-ITS2 rDNA. Further, the colonization of endophyte in the host plant by Light and Scanning Electron Microscopy will also be investigated. Whereas, genera Alternaria, Cochliobolus, Diaporthe, Epicoccum, Guignardia, Phoma, and Phomopsis, were identified; rDNA sequence analysis revealed intra-species variability among endophyte isolates of the genus Phomopsis sp. Light and Scanning Electron Microscopy techniques showed the presence of endophyte fungi inside L. divaricata leaves, inhabiting inter-and intra-cellular spaces. These types of extensive colonization and dissemination were reported throughout all the leaf parts in palisade parenchyma, esclerenchyma, spongy parenchyma, adaxial epidermis, and vascular bundle indicating colonization of endophytes in multiple structural sub-niches in the host plant.
ABSTRACT. Various types of organisms, mainly fungi and bacteria, live within vegetal organs and tissues, without causing damage to the plant. These microorganisms, which are called endophytes, can be useful for biological control and plant growth promotion; bioactive compounds from these organisms may have medical and pharmaceutical applications. Trichilia elegans (Meliaceae) is a native tree that grows abundantly in several regions of Brazil. Preparations using the leaves, seeds, bark, and roots of many species of the Meliaceae family have been widely used in traditional medicine, and some members of the Trichilia genus are used in Brazilian popular medicine. We assessed the diversity of endophytic fungi from two wild specimens of T. elegans, collected from a forest remnant, by sequencing ITS1-5.8S-ITS2 of rDNA of the isolates. The fungi were isolated and purified; 97 endophytic fungi were found; they were separated into 17 morpho-groups. Of the 97 endophytic fungi, four genera (Phomopsis, Diaporthe, Dothideomycete, and Cordyceps) with 11 morpho-groups were identified. Phomopsis was the most frequent genus among the identified endophytes. Phylogenetic analysis showed two major clades: Sordariomycetes, which includes three genera, Phomopsis, Diaporthe, and Cordyceps, and the clade Dothideomycetes, which was represented by the order Pleosporales.
ABSTRACT. The estuarine diatom Thalassiosira weissflogii (Fryxell & Hasle, 1977) has been widely used as live feed in aquaculture. The growth rate and biochemical composition of microalgae are highly influenced by environmental factors such as, light, salinity and nutrient availability. Salinity is difficult to control in some shrimp laboratories specialized in larvae production, because these laboratories depend upon the levels measured in estuaries or coastal lagoons, which are the water sources for larvae culture. The present study evaluated the effect of different salinities (25, 30, 35, 40, 45 and 50 psu), on the growth and chemical composition of T. weisflogii at three culture phases, under laboratory conditions. The highest growth rate and maximum cell density were found at 25 psu. Decrease in size and striking changes in morphology of the cells were observed at the higher salinities and drastic changes occurred at 50 psu. Protein and carbohydrate content were higher at low salinities (25 and 30 psu) during the stationary phase. The lipid production was higher at low salinities, but diminished as the phase changes occurred; in contrast, the lipid content was unaffected by the growth phase at higher salinities (≥35 psu). The higher growth rate and better biochemical composition were obtained at 25 and 30 psu. Keywords: estuarine diatom, microalgae culture, proximate composition. La diatomea estuarina Thalassiosira weissflogii (Fryxell & Hasle, 1977) ha sido utilizada como alimento vivo en acuacultura. La composición bioquímica del alimento vivo afecta la nutrición de los organismos durante su cultivo. La tasa de crecimiento y composición bioquímica de las microalgas están altamente influenciadas por factores ambientales como luz, salinidad y disponibilidad de nutrientes. En algunos laboratorios productores de larvas de camarón, es difícil controlar la salinidad, debido a que éstos dependen de los niveles presentes en estuarios o lagunas costeras, los cuales son la fuente de agua para el cultivo larvario. El presente estudio evaluó el efecto de diferentes salinidades (25, 30, 35, 40, 45 y 50 psu), sobre el crecimiento y la composición proximal de T. weissflogii en tres fases de cultivo, bajo condiciones de laboratorio. Las mayores tasas de crecimiento y la máxima densidad celular se obtuvieron a 25 psu. Se observó una reducción en tamaño y cambios en la morfología de las células a altas salinidades y los cambios drásticos ocurrieron a 50 psu. El contenido de proteínas y de carbohidratos fue más elevado a salinidades bajas (25 y 30 psu), durante la fase estacionaria de crecimiento. La producción de lípidos fue elevada a bajas salinidades y disminuyó a medida que cambiaba de fase; no se observó un efecto de las fases del cultivo sobre el contenido de lípidos en altas salinidades (≥35 psu).
