A multiyear survey of >200 wheat fields in Paraná (PR) and Rio Grande do Sul (RS) states was conducted to assess the extent and distribution of Fusarium graminearum species complex (FGSC) diversity in the southern Brazilian wheat agroecosystem. Five species and three trichothecene genotypes were found among 671 FGSC isolates from Fusarium head blight (FHB)-infected wheat heads: F. graminearum (83%) of the 15-acetyldeoxynivalenol (15-ADON) genotype, F. meridionale (12.8%) and F. asiaticum (0.4%) of the nivalenol (NIV) genotype, and F. cortaderiae (2.5%) and F. austroamericanum (0.9%) with either the NIV or the 3-ADON genotype. Regional differences in FGSC composition were observed, with F. meridionale and the NIV type being significantly (P<0.001) more prevalent in PR (>28%) than in RS (≤9%). Within RS, F. graminearum was overrepresented in fields below 600 m in elevation and in fields with higher levels of FHB incidence (P<0.05). Species composition was not significantly influenced by previous crop or the stage of grain development at sampling. Habitat-specific differences in FGSC composition were evaluated in three fields by characterizing a total of 189 isolates collected from corn stubble, air above the wheat canopy, and symptomatic wheat kernels. Significant differences in FGSC composition were observed among these habitats (P<0.001). Most strikingly, F. meridionale and F. cortaderiae of the NIV genotype accounted for the vast majority (>96%) of isolates from corn stubble, whereas F. graminearum with the 15-ADON genotype was dominant (>84%) among isolates from diseased wheat kernels. Potential differences in pathogenic fitness on wheat were also suggested by a greenhouse competitiveness assay in which F. graminearum was recovered at much higher frequency (>90%) than F. meridionale from four wheat varieties inoculated with an equal mixture of F. graminearum and F. meridionale isolates. Taken together, the data presented here suggest that FGSC composition and, consequently, the trichothecene contamination in wheat grown in southern Brazil is influenced by host adaptation and pathogenic fitness. Evidence that F. meridionale and F. cortaderiae with the NIV genotype are regionally significant contributors to FHB may have significant implications for food safety and the economics of cereal production.
A sample of 140 Fusarium graminearum isolates from Rio Grande do Sul, Brazil, representing three populations at least 150 km from one another, were examined for trichothecene genotype based on PCR amplification of portions of the Tri3 and Tri12 genes and a species-specific (Fg16F ⁄ R) primer pair. Genetic diversity was assessed in a sample of 103 F. graminearum lineage 7 (F. graminearum sensu stricto) isolates using amplified fragment length polymorphism (AFLP) markers. The 15-ADON genotype was dominant, followed by the NIV genotype (2-18% prevalence), across all three populations. All NIV-type isolates were in lineage 2 (F. meridionale) and all 15-ADON-type isolates were in lineage 7. Isolates with the same haplotype were rare and genotypic diversity was uniformly high ( ‡98% of the count), suggesting that recombination has played a significant role. The number of migrants (N m ) was estimated between 5 and 6 across all loci and all populations, but the high frequency of private alleles (up to 30%) suggests a historical, rather than contemporary, gene flow. Regarding linkage disequilibrium, 0AE8, 1AE5 and 2AE2% of the locus pairs from the three populations were in disequilibrium, which is lower than values reported in other locations. Thus, Brazilian populations differ from those found in Europe, North America and most of Asia in the presence of a significant frequency (7AE8%) of isolates of the NIV genotype in lineage 2.
O cultivo protegido tem sido um importante insumo agrícola que permite aumentos de produção das culturas, onde se esgotaram as tentativas convencionais de se obter incrementos face ao elevado emprego de técnicas modernas de cultivo. Nesse novo ambiente de cultivo, onde as plantas são colocadas sob novo limite de produtividade, visando propiciar condições para expressão do seu máximo potencial genético, o manejo inadequado dos seus fatores aéreos e do solo pode propiciar condições muito favoráveis a determinada doença biótica ou abiótica. Assim, doenças menos problemáticas ou de pouca importância em cultivo convencional, podem tornar-se muito destrutivas em cultivo protegido. Por isso, o manejo de doenças em cultivo protegido é uma tarefa complexa e medidas de controle devem ser integradas num sistema flexível, que seja compatível com o sistema de produção e que seja econômico. Desta forma, estratégias de manejo integrado das doenças em cultivo protegido podem ser agrupadas em medidas que visam a redução do inóculo inicial e aquelas que visam a redução da taxa de progresso da doença.

Com o objetivo de verificar o potencial de eucalipto (Eucalyptus citriodora) no controle alternativo de antracnose em pepino (Cucumus sativus), extrato aquoso (EA) desta essência florestal, autoclavado ou não autoclavado, nas concentrações de 0,1, 1, 5, 10, 15, 20 e 25% foi utilizado nos seguintes ensaios: indução de fitoalexinas em mesocótilos estiolados de sorgo (Sorghum bicolor) e em cotilédones de soja (Glycine max); fungitoxicidade in vitro sobre conídios de Colletotricum lagenarium; indução de resistência local ou sistêmica em plantas de pepino. Água e Bion foram utilizados como tratamentos controle. Os resultados indicaram que os EAs autoclavados e não autoclavados induziram a síntese de fitoalexinas em sorgo a partir da concentração de 1%. Em soja ocorreu síntese de gliceolina a partir de 10% e 15% dos EAs autoclavados e não autoclavados, respectivamente. Houve inibição total na germinação de esporos e formação de apressórios de C. lagenarium em concentrações de 20% e 1% do EA autoclavado, respectivamente. Para o extrato não autoclavado houve 75% de inibição da germinação de esporos em 25% do EA e inibição total da formação de apressórios em 15% do EA. Baseado no parâmetro tamanho de lesão, o extrato aquoso de E. citriodora, não autoclavado, apresenta potencial para induzir resistência local em pepino contra C. lagenarium.Palavras-chave adicionais: gliceolina, deoxiantocianidinas, resistência induzida.
