Background and Aims: Starting from the cytotoxic effects of monosodium glutamate (MSG), the aim of this study was to evaluate the protective effects of quince leaf extract as natural antioxidant on the reproductive dysfunction induced by monosodium glutamate in rats. Material and methods: Monosodium glutamate was administrated with a dose of 30 and 60 mg/kg and quince leaf extract was administrated with a dose of 500 mg/kg. At the end of study, body and testicular weight measurement, hormonal and epididymal sperm analysis were performed. Results: Follicle stimulating hormone (FSH) and testosterone levels were reduced after administration of monosodium glutamate. The levels of luteinizing hormone (LH) exhibited no significant changes. Treatment with quince leaf extract led to improvement in follicle stimulating hormone and testosterone levels. Epididymal sperm population was reduced after administration of monosodium glutamate and treatment with quince leaf extract. The increased sperm motility rate induced by monosodium glutamate was reduced after treatment with quince leaf extract. Administration of monosodium glutamate led to more body weight gain in comparison to combined administration monosodium glutamate and quince leaf extract. Conclusions: The quince leaf extract can be effective in reduction of functional alterations of reproductive system induced by monosodium glutamate.
Objective: Methylphenidate, Ritalin, is one of the most common medications. The administration of Ritalin leads to increase of the activity of central nervous system. Ritalin may be used for maintaining alertness and improving of attention which, may lead to increase of the risk of substance abuse in some cases. There is a little data about the effects of long term treatment with Ritalin on body organs involved in fertility ability. Regarding to the effect of normal fertility on the physical and mental health of males, this study was designed to investigate the time dependent effects of MPH on the activity of male reproductive system. Methods: Ritalin was administrated to adult rats (10 mg/kg/day) in three experimental groups for periods of two weeks, 11 weeks and 11 weeks with one week interval between each two weeks. The blood plasma levels of gonadotropins and testosterone was measured. Histomorphometrical study and sperm analysis was performed for evaluation of reproductive function. Results: The reduction in body weight and decline in the blood testosterone levels was seen in two groups which treated for 11 weeks. The reduction of microscopic indices of spermatogenesis and the alteration of morphometeric characteristics of seminiferous tubules was accompanied with epididymal sperm decrement in long term treated animals. Conclusion: The findings of this study indicate that, the long term use of methylphenidate can adversely damage the male fertility due to impairment of normal spermatogenesis. Background: Paclitaxel (PTX), a chemotherapeutic agent, and monosodium glutamate (MSG) have oxidative effects on testicular tissue.
Objective: In this study, the effects of MSG administration on the exacerbation of testicular tissue alterations related to PTX treatment were evaluated.
Materials and Methods: MSG (30 & 60 mg/kg i.p.) was administrated to six groups (n = 8/each) of adult mice before or after PTX treatment: control, PTX-treated, MSG30 + PTX, MSG60 + PTX, PTX + MSG30, and PTX + MSG60. Following the euthanizing, the body weight measurement, pituitary–testicular axis hormonal analysis and serum lipid peroxidation index assessment was prepared, testicular histomorphometry (tubular diameter and germinal epithelium height), immunohistochemistry of p53 was completed. Microscopic indices of spermatogenesis (tubular differentiation, spermiogenesis and repopulation indices) were studied.
Results: Body weight was not changed significantly. The levels of testosterone (p = 0.0001), follicle stimulating hormone (p = 0.019), and luteinizing hormone (p = 0.08) were decreased while the level of lipid peroxidation index was increased (p = 0.208) in the treated groups. The histomorphometry indices (p < 0.0001 and p = 0.001, respectively), germ cells population (p < 0.05) and microscopic indices of spermatogenesis (p = 0.001, p = 0.005, p < 0.0001, respectively) were significantly reduced in all treated groups. The administration of MSG before PTX treatment induces more changes. The most positive reaction to p53 was observed in MSG30 or 60 + PTX groups compared to other groups.
Conclusion: The administration of MSG could intensify testicular tissue alterations related to PTX chemotherapy.
