Red sugarcane (RS) is a lesser bred of sugarcane variant in Malaysia compared to yellow sugarcane (YS), thus less data is available regarding this species. On this study, the physicochemical and microbiological properties of RS was determined and compared with YS under different conditions. Both sugarcane variants were subjected to heat and sonication treatments for 5 and 15 minutes as a method for preservation. RS contains a higher °Brix (15) compared to YS (14), and was unchanged while YS demonstrated a significant reduction. Furthermore, RS displayed a superior fructose (+802%), glucose (+119%) and sucrose (+17.91%) levels in all samples and they were largely preserved during both treatments. However, RS was sensitive to colour changes, as darker juice (decrease in lightness and increase in redness) was produced after treatments. RS possessed lower phenolic content (-31.15%) and antioxidant activity (-11.51%) than YS, and was significantly affected by both treatments. Sonication was more effective in reducing the microbial content than heat in both treatments. These findings indicated that RS is better at retaining its physicochemical properties under preservation treatment, but less resistant in preserving its appearance and nutritional values.
Purpose: This study aimed to assess the effect of nitrogen, salt and pre-culture conditions on the production of lovastatin in A. terreus ATCC 20542. Methods: Different combinations of nitrogen sources, salts and pre-culture combinations were applied in the fermentation media and lovastatin yield was analysed chromatographically. Result: The exclusion of MnSO₄ •5H₂O, CuSO₄•5H₂O and FeCl₃•6H₂O were shown to significantly improve lovastatin production (282%), while KH2PO₄, MgSO₄•7H₂O, and NaCl and ZnSO4•7H₂O were indispensable for good lovastatin production. Simple nitrogen source (ammonia) was unfavourable for morphology, growth and lovastatin production. In contrast, yeast extract (complex nitrogen source) produced the highest lovastatin yield (25.52 mg/L), while powdered soybean favoured the production of co-metabolites ((+)-geodin and sulochrin). Intermediate lactose: yeast extract (5:4) ratio produced the optimal lovastatin yield (12.33 mg/L) during preculture, while high (5:2) or low (5:6) lactose to yeast extract ratio produced significantly lower lovastatin yield (7.98 mg/L and 9.12 mg/L, respectively). High spore concentration, up to 107 spores/L was shown to be beneficial for lovastatin, but not for co-metabolite production, while higher spore age was shown to be beneficial for all of its metabolites. Conclusion: The findings from these investigations could be used for future cultivation of A. terreus in the production of desired metabolites.
The intensive amount of chemical usage in agricultural practices could contribute to a significant impact on food safety issues and environmental health. Over-usage of chemical fertilisers may alter soil characteristics and contaminate water sources, leading to several human and animal health issues. Recently, there have been efforts to use microbial biofertilisers as a more sustainable and environmentally friendly agricultural practice in the common household of Southeast Asia. Traditionally, this method tends to utilise leftover food materials and readily available bacterial cultures, such as yoghurt drinks, and ferment them under a specific period in either solid or liquid form. So far, most of the testimonial-based feedbacks from local communities have been positive, but only limited information is available in the literature regarding the usage of biofertiliser fermented food (BFF). Previously, raw food waste has been used in the agriculture system to promote plant growth, however, the functional role of fermented food in enhancing plant growth have yet to be discovered. An understanding of the symbiotic relationship between fermented food and plants could be exploited to improve agricultural plant production more sustainably. Fermented food is known to be rich in good microbial flora (especially lactic acid bacteria (LAB)). LAB exist in different sources of fermented food and can act as a plant growth-promoting agent, improving the nutrient availability of food waste and other organic materials. Therefore, in this review, the potential use of seafood-based, plant-based, and animal-based fermented food as biofertiliser, especially from Southeast Asia, will be discussed based on their types and microbial and nutritional contents. The different types of fermented food provide a wide range of microbial flora for the enrichment of proteins, amino acids, vitamins, and minerals content in enhancing plant growth and overall development of the plant. The current advances of biofertiliser and practices of BFF will also be discussed in this review.
Crude glycerol (CG) can be used as a substrate for microbial bioconversion. However, due to presence of many impurities, many microorganisms are unable to utilise this substrate efficiently. The present study is trying to improve CG using as the feedstock of Aspergillus terreus for the production of lovastatin, (+)-geodin, and sulochrin. The CG was pretreated chemically (solvents) and physically (activated carbon (AC) and water softener (WS)) to separate most of the impurities from the CG. For solvent pretreatments, petroleum ether (PE) produced the largest increase of lovastatin (92.8%) when compared to positive control and pure glycerol (PG) and up to 820% when compared to negative control (CG). In contrast, diethyl ether (DE) produced the largest increase in (+)-geodin at 80.81% (versus CG) and 176.23% (versus PG). The largest increase in toluene (Tol) was observed in sulochrin production, at 67.22% (versus CG) and 183.85% (versus PG). For physical pretreatments, the pattern of metabolite production in AC (lovastatin: 20.65 mg/L, (+)-geodin: 7.42 mg/L, sulochrin: 11.74 mg/L) resembled PG (lovastatin: 21.8 mg/L, (+)-geodin: 8.60 mg/L, sulochrin: 8.18 mg/L), while WS (lovastatin: 11.25 mg/L, (+)-geodin: 15.38 mg/L, sulochrin: 16.85 mg/L) resembled CG (lovastatin: 7.1 mg/L, (+)-geodin: 17.10 mg/L, sulochrin: 14.78 mg/L) at day 6 of fermentation. These results indicate that solvent pretreatments on CG are excellent for metabolites production in A. terreus, depending on the solvents used. In contrast, physical pretreatments are only feasible for (+)-geodin and sulochrin production. Therefore, different strategies can be employed to manipulate the A. terreus bioconversion using improved CG by using a few simple pretreatment strategies.
