Menopause is a natural phase of life and happens in every woman who reaches a certain age. This condition has many problems, such as endometrial atrophy, which can cause abnormal bleeding. Sauropus androgynus, known as Katu Leaf, has isoflavones compound and acts as an estrogen-like agent or phytoestrogen. Isoflavone can induce cell proliferation activity in increasing endometrial thickness and uterine weight. This study was aimed to find how the is flavone compound in Sauropus androgynus can increase the uterine weight in menopausal mice as an animal model. Twenty female mice aged two months and one week were given VCD (4-Vinyl cyclohexane dioxide) to induce the atresia acceleration of ovarian follicles, then treated with Katu Leaf extract of 0, 15 and 30mg/kgBW over four weeks. The uterine weights were measured and analyzed with One Way Analysis of Variances (Anova) and further tested with Least Significance Difference in 95% confidence interval. The result showed a difference between control and treatments group (p<0.05) with 30mg/kgBW as highest dosage, which increase uterine weight as 112.80 +8.86mg), but this is not higher than normal (116.36+6.83mg). This study suggests that Katu Leaf extract has an isoflavone compound which can increase uterine weight in menopausal mice.
Preeclampsia is the worldwide leading cause of fetomaternal morbidity and mortality which involves the placental dysfunction. A poor placentation and formed of non-dilated spiral artery caused utero-placental circulation insufficiency, resulted in inadequate supply of nutrients and oxygen to support normal aerobic growth of the fetus. Apium graveolens or celery has been widely known as antioxidant, antiinflammation and antihypertensive with flavonoid-apigenin as main active compound. Apigenin can inhibit TNF-α, HIF-1α and nitric oxide blocking as major pathophysiological pathway of preeclampsia. This study was aimed to find how the Apium graveolens can improve intrauterine growth and its correlation with suppression of anti-angiogenic factor sFlt-1 in anti-Qa2 preeclampsia animal model. Twenty female BALB/c Mus musculus were divided into 4 groups: control, anti-Qa2 and anti-Qa2 with 500 and 1000 mg/kgBW celery herbs extract. The fetal weights and lengths, placental weights and serum sFlt-1 levels were measured and analyzed with One Way ANOVA and further tested with Least Significance Difference in 95% confidence interval. The result showed a difference between control and treatments group (p≤0.05) with 1000 mg/kgBW significantly increase intrauterine growth and decrease sFlt-1, then there is a negative correlation between intrauterine weight and serum sFlt-1. This study suggests that celery herbs extract (CHE) has an apigenin-flavonoid compound which can prevent intrauterine growth restriction (IUGR) via suppression of antiangiogenic factor production in preeclampsia mice model.
Objective: To examine the potency of estrogenic effect of Katuk (or star gooseberry or sweet leaf) Sauropus androgynus L. Merr (Phyllanthaceae) leaf extract on the vaginal and endometrial atrophy in perimenopausal mice model. Methods: Completely random experiments divided into 4 groups with 5 female mice in each group: control group (N), perimenopause group which induced by 4-vynil cyclohexene dioxide (VCD) (P0), perimenopause group followed by S. androgynus leaves extract administration with 15 and 30 mg/kg BW (P15 and P30). The mice were induced to artificial perimenopause with 160 mg/kg body weight (BW) of VCD 5 times/week for 2 weeks and continuously treated with S. androgynus leaf extract for 30 days. Vaginal smear examination, maturation index (MI), vaginal epithelium and endometrial thickness were observed. Data were analyzed using a standard one-way ANOVA with LSD Test by SPSS 16.0 for Windows. Results: Administration of S. androgynus leaf extract for 30 days to perimenopausal mice model significantly decreased the duration of mice diestrus phase (p<0.01) compared to perimenopause mice. S. androgynus leaf extract treatment at dose 30 mg/kg BW led to higher vaginal epithelium thickness and maturation index compared to another group (p<0.01). The effective dose to enhance the vaginal epithelial and endometrial cells proliferation was 30 mg/ kg BW. Conclusions: S. androgynus leaf extract had a good estrogenic action that it might be useful for therapy of vaginal and endometrial atrophy caused by perimenopause effect.
