Our results indicate that 34 % of the variance in the required propofol dose may be explained by these factors and that CYP2B6 c.516G>T polymorphism, which decreases the metabolism of the drug, accounts for approximately 7 % of the drug dosage. Our results show the possible influence of CYP2B6 c.516G>T genetic variant on propofol dose in patients under general anaesthesia.
Obesity is associated with insulin resistance (IR) and increased oxidative stress. Thus, the present study aimed to evaluate anthropometric parameters, IR, and oxidative stress in obese individuals subjected to two types of concurrent training at the same intensity but differing in frequency. Accordingly, 25 individuals were divided into two groups: concurrent training 1 (CT1) (5 d/wk) and concurrent training 2 (CT2) (3 d/wk), both with moderate intensity. Anthropometric parameters, IR, and oxidative stress were analyzed before and after 26 sessions of training. Both groups had reduced body weight and body mass index (P < 0.05), but only CT1 showed lower body fat percentage and increased basal metabolic rate (P < 0.05). Moreover, CT1 had increased HOMA-IR and decreased protein damage (carbonyl level), and CT2 had decreased HOMA-IR and increased lipid peroxidation (TBARS level) (P < 0.05). On the other hand, both training protocols reduced the GPx activity. It can be concluded that both types of concurrent training could be an alternative for lowering body weight and BMI. Also, it was observed that concurrent training, depending on the frequency, can contribute to reducing body fat, oxidative damage (protein oxidation), and IR but can induce oxidative damage to lipids. More studies are needed to elucidate the mechanisms involved.
The number of people who are overweight or obese is increasing worldwide and the quality of life of these people can be affected by their condition. Physical training has been studied in obese patients and is correlated with low-grade inflammation and alterations in the immune system. This study investigated the effect of concurrent training on anthropometric, inflammatory, and immunological parameters in overweight and obese adults. Fourteen sedentary volunteers (men and women) with a body mass index between 25 kg/m 2 and 39.9 kg/m 2 from Porto Alegre, Brazil attended a 12-week course of concurrent training. We analyzed: prior to and after training, anthropometric parameters, cytokine serum levels (interleukin 6, tumor necrosis factor a, interferon g, interleukin 17A and interleukin 10; measured by enzyme-linked immunosorbent assay), high-sensitive C reactive protein (measured by turbidimetry), and the frequency of T lymphocytes and monocytes (CD3þCD4þ, CD3þCD8þ, and HLA-DRþ) in peripheral blood (measured by flow cytometry). The sample consisted of ten women and four men with a mean ± SD age of 47.58 ± 3.01 years. After 12 weeks of training we observed a reduction in body weight, body mass index, waist, abdomen, and hip circumferences, the percentage mass of fat, and an increase in the time taken to reach exhaustion (p < 0.05). The participants had increased frequencies of CD3þCD4þ and CD3þCD8þ T lymphocytes and a reduction in the frequencies of HLA-DRþ monocytes (p < 0.05). Interestingly, the levels of tumor necrosis factor a and high-sensitive C reactive protein increased (p < 0.05). Our data suggest that concurrent training can improve body composition as well as increasing T cell proliferation in overweight and obese patients. However, the progression of the exercises can be physiologically stressful to these patients, as demonstrated by the inflammatory markers.
A obesidade é uma doença crônica e multifatorial, que vem aumentando sua incidência ao longo dos anos e está sendo considerada um problema de saúde pública. É caracterizada por um estado crônico de baixo grau de inflamação, no qual os níveis de proteína C reativa geralmente estão aumentados. Esse processo inflamatório pode alterar a função endotelial nesses indivíduos, uma vez que os níveis elevados de proteína C reativa estão relacionados a uma baixa produção de óxido nítrico, o qual é responsável pela vasodilatação do endotélio. Vários estudos têm demonstrado a influência de polimorfismos nos genes da proteína C reativa e da enzima óxido nítrico sintase endotelial em indivíduos obesos e saudáveis. Dessa forma, esta revisão de literatura tem como objetivo discutir a relação da obesidade com níveis de PCR e função endotelial, bem como elucidar a associação de fatores genéticos. Foram consultados artigos científicos dos bancos de dados PubMed, Science Direct e Scielo, utilizando os seguintes descritores e combinações entre eles: proteína C reativa, função/ disfunção endotelial, polimorfismo e obesidade e seus respectivos em inglês. Com isso, concluímos que indivíduos obesos apresentam elevados níveis de proteína C reativa, em decorrência do excesso de gordura corporal, e isso implica uma redução na síntese de óxido nítrico. Todos esses fatores podem ser influenciados por polimorfismos nos genes reguladores da produção dessas proteínas.
