Objectives We aimed to determine the differences between normozoospermic and oligozoospermic individuals according to levels of spermatid-specific thioredoxin reductase 3 (SPTRXR3/STRX3/TXNDC8/TXNRD3) and testis expressed protein 101 (TEX-101), and to evaluate the correlations between spermiogram data and biochemical parameters. Methods The study was carried out at the Andrology Laboratory of Medicine Faculty of Selcuk University. Two groups were designed: Group 1: Normozoospermia (n=40, sperm concentration ≥ 15 million/mL), Group 2: Oligozoospermia; (n=40, sperm concentration < 15 million/mL). Seminal plasma SPTRXR3 and TEX-101 levels were analyzed with ELISA method. Spermiogram analysis was evaluated according to WHO 2010 Kruger criteria. Results TEX-101 protein levels were significantly different in normozoospermia (2.12 ± 0.08 ng/mL) compared to oligozoospermia (1.55 ± 0.04 ng/mL). SPTRXR3 levels (6.98 ± 0.46 ng/mL) were higher in oligozoospermia than normozoospermia (3.07 ± 0.35 ng/mL). Both TEX-101 and SPTRXR3 levels were correlated statistically with most of the spermiogram parameters. Conclusions High SPTRXR3 and low TEX-101 levels may be a biomarker in evaluation of male infertility. The relations between spermiogram parameters indicates that results present a new clinical approach in biology of oligozoospermic male.
Aim: In this study, it was aimed to determine the total phenolic content, flavonoid content, and antioxidant activity of Castanea sativa Mill. (chestnut) leaf extract and their anti-proliferative effect on MCF 7 cell line. Materials and Methods: It determined antioxidant capacities by using DPPH assay, total phenolic content by using Folin-Ciocaltaeu method and Total flavonoid content by aluminum chloride colorimetric method in the ethanolic extracts of Chesnut leaf. In addition, the anti-proliferative effect of chestnut leaf extract was determined by XTT method in MCF7 breast cell line. The leaf extract used was applied at different concentrations for 24 hours, 48 hours and 72 hours and the results were evaluated with the Graphpad Prism software program. Results: In this study, it was revealed that the total phenolic contents of ethanolic extracts of chestnut leaf 58,22 mg GAE/g. Total flavonoid content was 64,62 mg QE/g. The DPPH activity of the leaf extract of chestnut was 80.06%. Moreover, findings showed that Chestnut leaf extract had cytotoxic effect against breast cancer cells depends on concentration and time. The 24h, 48h and 72h most effective IC50 dose were 100.1 μL ,193 μL 15.23 μL, respectively. Conclusion: Chestnut has potential antiproliferative and antioxidant activities in conjunction with its high phenolic and flavonoid content on the MCF-7 cell line. However, more studies are needed.
ÖZETİnfertiliteden etkilenen çiftler giderek artmaktadır. Erkek infertilite değerlendirilmesinde ilk adım, semen analizidir. Ancak seminal kompozisyon çevresel faktörlerden ve diğer patolojik durumlardan etkilendiği için erkek infertilite tanısında kesin bir sonuç vermediği durumlar söz konusudur. Bu nedenledir ki, erkek infertilitesinin tanısı veya tedavisi sürecinde farklı disiplinlerin araştırdığı diagnostik ve prognostik testlere ihtiyaç duyulmakta ve son yıllarda artan ivme ile çalışmalar devam etmektedir. Seminal plazma sıklıkla biyoloji alanının fertilizasyon durumunun değerlendirilmesinde tercih ettiği numune tipidir. Seminal plazmada kolay analiz edilebilen, biyokimyasal açıdan test duyarlılığı ve özgüllüğü yüksek biyobelirteçlerin belirlenmesi ve tanımlanmasının spermiyogram analizlerine ilaveten tanı ve tedavide infertil erkeklerin daha iyi tanımlanmasında bir yöntem olarak kullanılabilir. Dolayısıyla seminal plazma biyobelirteçleri ilerleyen zamanlarda erkek faktörlü infertilitenin değerlendirilmesinde ön analizlerden olacak gibi görünmektedir. Güncel çalışmalar seminal plazma biyobelirteçlerinin, azospermi vakalarında invaziv testis biyopsisine ek olarak yapılabileceğini ve hatta bazı belirteçlerin öncelikli olarak tercih edilebileceğini göstermektedir. Bununla birlikte, obstrüktif ve non-obstrüktif azospermi ayrımının yapılabildiği bildirilmektedir. Bununla birlikte, infertil erkek bireylerde yakın gelecekte spermiyogram analizlerinin yanında diagnostik ve prognostik biyobelirteçlerin biyokimyasal rollerini ve analizlerinin önemini vurgulamak üzere planlanan bu derlemenin literatüre katkı sağlayacağını düşünmekteyiz.
