Pyrosequencing of 16S rRNA gene amplicons for microbial community profiling can, for equivalent costs, yield more than two orders of magnitude more sensitivity than traditional PCR cloning and Sanger sequencing. With this increased sensitivity and the ability to analyze multiple samples in parallel, it has become possible to evaluate several technical aspects of PCR-based community structure profiling methods. We tested the effect of amplicon length and primer pair on estimates of species richness (number of species) and evenness (relative abundance of species) by assessing the potentially tractable microbial community residing in the termite hindgut. Two regions of the 16S rRNA gene were sequenced from one of two common priming sites, spanning the V1-V2 or V8 regions, using amplicons ranging in length from 352 to 1443 bp. Our results show that both amplicon length and primer pair markedly influence estimates of richness and evenness. However, estimates of species evenness are consistent among different primer pairs targeting the same region. These results highlight the importance of experimental methodology when comparing diversity estimates across communities.
This paper presents a methodology for regional parameter estimation of the three-layer Variable Infiltration Capacity (VIC-3L) land surface model with the goal of improving the streamflow simulation for river basins in China. This methodology is designed to obtain model parameter estimates from a limited number of calibrated basins and then regionalize them to uncalibrated basins based on climate characteristics and large river basin domains, and ultimately to continental China. Fourteen basins from different climatic zones and large river basins were chosen for model calibration. For each of these basins, seven runoff-related model parameters were calibrated using a systematic manual calibration approach. These calibrated parameters were then transferred within the climate and large river basin zones or climatic zones to the uncalibrated basins. To test the efficiency of the parameter regionalization method, a verification study was conducted on 19 independent river basins in China. Overall, the regionalized parameters, when evaluated against the a priori parameter estimates, were able to reduce the model bias by 0.4%-249.8% and relative root-mean-squared error by 0.2%-119.1% and increase the Nash-Sutcliffe efficiency of the streamflow simulation by 1.9%-31.7% for most of the tested basins. The transferred parameters were then used to perform a hydrological simulation over all of China so as to test the applicability of the regionalized parameters on a continental scale. The continental simulation results agree well with the observations at regional scales, indicating that the tested regionalization method is a promising scheme for parameter estimation for ungauged basins in China.
Inhibitory effects of the Noble muscadine grape extracts and the representative phytochemicals for anthocyanins (i.e., cyanidin and cyanidin-3,5-diglucoside) on two enzymes, that is, α-glucosidase and pancreatic lipase, were investigated regarding their antidiabetic activities. The study demonstrated that the anthocyanin extracts and the selected chemicals obeyed the competitive mode against the enzymes. The methanolic extracts of whole fruit and skin of the muscadine showed inhibitory activities against the α-glucosidase with their IC(50) values at 1.50 and 2.73 mg/mL, and those against the lipase at 16.90 and 11.15 mg/mL, respectively, which indicated that the muscadine extracts possessed strong antidiabetic activities. Particularly, the ethyl acetate (EtoAc) extract and the butanol (BuOH) extract exhibited much higher inhibitory activities against both enzymes than the CHCl(3) and water extracts, while the majority of anthocyanins existed in the BuOH fractions. Moreover, cyanidin exhibited a much stronger antidiabetic activity than cyanidin-3,5-diglucoside, suggesting that anthocyanins may have higher inhibitory activities after being digested. Further chromatographic analysis by high-performance liquid chromatography-mass spectrometry identified five individual anthocyanins, including cyanidin, delphinidin, petunidin, peonidin, and malvidin glycosides.
