Intramuscular fat (IMF) content and fatty acid (FA) composition vary significantly across beef cattle breeds, which play an important role in taste and nutritional value. However, the molecular mechanisms underlying these phenotypic differences remain unknown. The present study compared meat quality traits between Yunling cattle and Chinese Simmental cattle. Yunling cattle showed a lower IMF content and proportion of monounsaturated fatty acids (MUFA), as well as higher proportions of saturated fatty acids (SFA), polyunsaturated fatty acids (PUFA), and short-chain fatty acids (sc-FA) in the longissimus dorsi (LD) muscle than Chinese Simmental cattle. To further identify the candidate genes and pathways responsible for these phenotypic differences, the transcriptome of LD muscle from the two breeds were measured using RNA-seq. A total of 1347 differentially expressed genes were identified. The major metabolic pathways that were differentially modulated were lipolysis and glycometabolism. Yunling cattle showed a higher expression of lipolysis genes (ALDH9A1, ACSL5, ACADM, ACAT2, ACOT2) and a lower expression of genes related to glycometabolism (PGM1, GALM, PGM1, GPI, LDHA). This research identified candidate genes and pathways for IMF content and FA composition in the LD muscle of beef cattle, which may facilitate the design of new selection strategies to improve meat quality.
In part as a result of the production of an enterotoxin, Staphylococcus aureus is a highly infectious pathogen and is a considerable threat to food hygiene and safety. Clinical mastitis models were established by S. aureus nipple-tube perfusion. The influence of mastitis on the mammary-gland-tissue proteomic profile was investigated using isobaric tags for relative and absolute quantitation. In this study, healthy and mastitic tissues from different mammary-gland areas of the same dairy cows were screened, and differentially expressed proteins were identified. Bioinformatic analysis identified proteins related to the inflammation and immunization of dairy cows. Histology, immunoblotting, and immunohistochemical-staining analyses were used to determine the expression of PGLYRP1 and PTX3 proteins in the acquired mammary-gland-tissue samples. PGLYRP1 and PTX3 in mastitic mammary glands may be associated with tissue damage and immune responses to late stages of infection. This further contributes to the understanding of the molecular theory of the treatment of mastitis caused by S. aureus.
In this research paper we filter and verify miRNAs which may target silent information regulator homolog 2 (SIRT2) gene and then describe the mechanism whereby miRNA-212 might regulate lipogenic genes in mammary epithelial cell lines via targeting SIRT2. Bioinformatics analysis revealed that the bovine SIRT2 gene is regulated by three miRNAs: miR-212, miR-375 and miR-655. The three miRNAs were verified and screened by qRT-PCR, western blot, and luciferase multiplex verification techniques and only miR-212 was shown to have a targeting relationship with SIRT2. The results of co-transfecting miR-212 and silencing RNA (siRNA) showed that by targeting SIRT2, miR-212 can regulate the expression of fatty acid synthetase (FASN) and sterol regulatory element binding factor 1 (SREBP1) but not peroxisome proliferator-activated receptor gamma (PPARγ). Measurement of triglyceride (TAG) content showed that miR-212 increased the fat content of mammary epithelial cell lines. The study indicates that miR-212 could target and inhibit the expression of the SIRT2 gene to promote lipogenesis in mammary epithelial cell lines.
In order to investigate the factors affecting milk urea nitrogen in Chinese Holstein cows, a large commercial dairy farm participated in a 30-month study. In this study, the mean milk urea nitrogen concentration was 11.75 mg/dl. The milk urea nitrogen reached its maximum value on day 90 of lactation for the first parity and the third or higher parities, but it peaked at the end of lactation for the second parity. The milk urea nitrogen of the first parity was lower than that of other parities. The milk urea nitrogen showed its minimum level in January, and reached its maximum in July. The milk urea nitrogen at the first month of lactation in cows calving in summer was higher than other seasons, while at the fourth month of lactation, the milk urea nitrogen of cows calving in autumn was significantly lower than in cows calving in other seasons. Positive correlations were observed between daily milk yield, net energy for lactation, crude protein and milk urea nitrogen for the first and third parities, but negative correlations were observed in the second parity. The milk urea nitrogen showed significantly positive correlations with fat content, total solid content and daily matter intake for all parities. A negative correlation was observed between milk urea nitrogen and protein content, with the exception of the second parity. For all data, as milk urea nitrogen concentration increased, milk protein content decreased. It has been recommended that milk urea nitrogen concentration should be evaluated in combination with parity, days in milk, season (or month), daily matter intake and dietary nutritional components, in order to improve the management and economic benefits of dairy farm.
