Samples from three different mating stages (before, during and after mating) of the Mediterranean fruit fly Ceratitis capitata were used in this experiment. Samples obtained from whole insects were subjected to extraction with the two mixtures of solvents (acetonitrile/water (A) and methanol/acetonitrile/water (B)) and a comparative study of the extractions using the different solvents was performed. Direct immersion-solid phase microextraction (DI-SPME) was employed, followed by gas chromatographic-mass spectrometry analyses (GC/MS) for the collection, separation and identification of compounds. The method was validated by testing its sensitivity, linearity and reproducibility. The main compounds identified in the three different mating stages were ethyl glycolate, α-farnesene, decanoic acid octyl ester, 2,6,10,15-tetramethylheptadecane, 11-tricosene, 9,12-(Z,Z)-octadecadienoic acid, methyl stearate, 9-(Z)-tricosene, 9,11-didehydro-lumisterol acetate; 1,54-dibromotetrapentacontane, 9-(Z)-hexadecenoic acid hexadecyl ester, 9-(E)-octadecenoic acid and 9-(Z)-hexadecenoic acid octadecyl ester. The novel findings indicated that compound compositions were not significantly different before and during mating. However, new chemical compounds were generated after mating, such as 1-iodododecane, 9-(Z)-tricosene and 11,13-dimethyl-12-tetradecen-1-acetate which were extracted with both (A) and (B) and dodecanoic acid, (Z)-oleic acid, octadecanoic acid and hentriacontane which were extracted with (A) and ethyl glycolate, 9-hexadecenoic acid hexadecyl ester, palmitoleic acid and 9-(E)-octadecenoic acid, which were extracted with solvent (B). This study has demonstrated that DI-SPME is useful in quantitative insect metabolomics by determining changes in the metabolic compounds in response to mating periods. DI-SPME chemical extraction technology might offer analysis of metabolites that could potentially enhance our understanding on the evolution of the medfly.
Timely detection of Mediterranean fruit fly (Medfly) is very important so that eradication action can be taken on time. The larvae stage of this insect is the most dangerous stage as it is within the pulp of the fruit, making it hard to detect by visual inspection. In most countries at ports of entry the inspector check a small sample of fruit by visual inspection or by cutting the produce and searching for fungus and pests. This paper will investigate a quick, reliable and sensitive method to determine the presence of fruit flies. Our research focuses on developing the technology for detecting hidden infestations by using the Head Space-Soild Phase Micro Extraction (HS-SPME) method coupled with Gas Chromatography-Mass Spectrocopy (GC-MS) technique. Five different types of fruit were infested with an early stage of Medfly Ceratitis capitata Wiedemann (Diptera: Tephidae). We investigated to detect the differences in volatile organic compounds (VOC's) between infested and noninfested fruits by using HS-SPME with (GC-MS). The results indicated that for few chemicals no significant differences between infested and non-infested fruit can be seen, especially in the fruits with first instar. However, in case of third instar larvae infested fruits significant differences in the chemicals can be seen as compare to non infested fruits and other instar infestations. These chemicals include ethyl (Z)-2 butenoate, 2-heptanone, anisole, β-cis-ocimene, 1,3,7-nonatriene,4,8-dimethy-,ethyl octyate, isoamyl caproate and 1β,4βh,10βh-guaia-5,11-diene, in apple. Ethyl (Z)-2-butenoate, (+)-2-bornanone, (-)-transisopiperitenol, methyl caprate, caryophyllene and farnesene in orange. Butanoic acid, 3-methyl-,2-methylbutul acetate, sabinene, β-myrcene, octanoic acid, methyl ester, dihydrocarvone, (-)-trans-isopiperitenol and ethyl laurate in mandarin. Butyl 2-methylbutanoate, terpinen-4-ol, P-menth-8-en-2-one, 155E-,(3E,7E)-4,8,12-trimethyltrideca-1,3,7,11-tetraene and dodecanoic acid, ethyl ester in lemon. Decane, 3-methyl-, p-menth-1,4(8)-diene, 1-undecene and α-cubebene in avocado. Thus, the VOC's method could provide a possible tool for detecting tephritid larvae and this method could be adopted by industries importing and exporting fruit.
Antifungal potential of leaves and roots parts of Chenopodium album was evaluated against five phytopathogenic fungi including Alternaria alternata, Fusarium solani, Rhizoctonia solani, Pythium aphanidermatum and Sclerotinia sclerotium using four concentrations viz. 0, 5, 10 and 15% of leaves and roots water extract led to significantly reduced of fungi mycelial growth. GC-MS analysis of leaves and roots water extract showed the presence of 7 compounds. 2(3H)-furanone, dihydro-4,4-dimethyl; 9-octadecenoic acid (Z), methyl ester; 9,12-octadecenoic acid (Z), methyl ester; 6-methylene bicyclo (3.2.0) hept-3-en-2-one., 1,2-benzene dicarboxylic acid, mono (2-ethylhexyl) ester and hexadecanoic acid, methyl ester. Five phytopathogenic fungi could be managed and controlled by the water leaves and roots extract of C. album. The antifungal activity of this extract was possible could be used to control a wide range of fungi.
