Biofilm formation is considered to be a selective advantage for Staphylococcus aureus mastitis isolates by facilitating bacterial persistence in the udder. It requires attachment to mammary epithelium, proliferation and accumulation of cells in multilayers. The objective of this study was to determine the sensitivity and specificity of three techniques for the detection of S. aureus biofilm-positive strains. Two phenotypic tests, including growth on microtitre plates and Congo red agar, were compared with a PCR technique using 94 S. aureus strains obtained from cows with subclinical mastitis from two farms in the state of São Paulo. These strains were characterised by in vitro slime production on Congo red agar, biofilm formation on microtitre plates and the presence of the icaA and icaD genes. The results revealed that 85% of the isolates tested produced slime on the Congo red agar, 98.9% of the isolates produced biofilms in vitro by adhering to sterile 96-well “U” bottom polystyrene tissue culture plates, and 95.7% of the isolates carried the icaA and icaD genes. The results of the phenotypic tests for biofilm formation were compared with those of the molecular analysis, and the sensitivity and specificity of the Congo red agar test were 88.9% and 100%, respectively, while those of the microtitre plate test were 100% and 25%, respectively. When the phenotypic methods for the detection of biofilm producers, namely growth on microtitre plates and Congo red agar, were compared, the sensitivity and specificity were 86% and 100%, respectively. Therefore, growth on Congo red agar and the microtitre plate test are methods that could be used to determine whether an isolate has the potential for biofilm production.
The objective of this study was to determine the prevalence of Shigatoxigenic Escherichia coli (STEC) and STEC serogroups O157, O111 and O113 in feces, water and milk sampled in dairy farms in Jaboticabal, SP, Brazil. Feces (n=454), water (n=54) and milk samples (n=30) were collected from 10 herds and assessed for the presence of the virulence genes stx 1 , stx 2 and eae by polymerase chain reaction (PCR). All stx and eae positive samples were submitted to a second PCR reaction targeting the sequences rfb O157, rfb O111 and rfb O113. High prevalence of stx was detected (59.9%) in fecal samples, whereas the prevalence of sequences rfb O157, rfb O111 and rfb O113 was 18.9%, 3.3% and 30.4%, respectively. All sequences were detected more frequently in calves and heifers. Sequences stx2 and eae were prevalent in the fecal samples. stx sequences were detected in 1.9% and 3.3% of water and milk samples, respectively. Low prevalence of E. coli O113 was observed in water samples, whereas no E. coli O157 or E. coli O111 was detected. Furthermore, none of the serogroups were identified in milk samples. STEC was identified in all herds (100%), and serogroups O157, O111 and O113 were observed in 40%, 50% and 90% of the herds, respectively. In conclusion, the high STEC prevalence detected in dairy herds evidences that bovine feces might play an important role as a contamination source in the region of Jaboticabal, Brazil.
RESUMO Este trabalho teve como objetivo determinar a prevalência de Escherichia coli produtoras de Shigatoxinas (STEC) e E. coli dos sorogrupos O157, O111 e O113 em rebanhos leiteiros do Município de Jaboticabal, SP, Brasil. A presença de sequências gênicas stx1, stx2e eae foi detectada pela reação em cadeia da polimerase (PCR) em amostras de fezes. Todas as amostras stx e eae positivas foram submetidas a uma nova reação de PCR para detecção das sequências rfb O157, O111 e O113. Observou-se uma alta prevalência (72,16%) de sequências stx nas fezes dos bovinos, sendo que o perfil genotípico encontrado com maior frequência foi o stx1 associado à stx2. Os coeficientes de prevalência das sequências rfb O157, O111 e O113 foram, respectivamente, 14,77%, 0,2% e 30,83%. Animais de todos os rebanhos (100%) apresentaram em suas fezes STEC e E. coli O113 e os sorogrupos O157 e O111 foram observados em 60,0% e 10,0% dos rebanhos, respectivamente. Concluiu-se que a alta prevalência de STEC detectada em rebanho leiteiro evidenciada nas fezes de bovinos desempenham um papel importante na contaminação ambiental e podem oferecerrisco de agravo à saúde pública.
