Pengelolaan dan pelestarian keanekaragaman hayati berawal dari pengetahuan tentang ketersediaan sumberdaya, identifikasi, dan sistematika spesies. Banyaknya spesies Turbinidae dengan berbagai bentuk morfologi dan transformasi bentuk menjadikan identifikasi dengan gen COI sangat penting dalam mempercepat pengungkapan identitas spesies. Penelitian ini dilakukan dengan tujuan menganalisis pola pertumbuhan dan mengidentifikasi gastopoda Turbinidae dengan pendekatan DNA barcode. Hasil identifikasi berdasarkan karakter morfologi menunjukkan bahwa genus Turbo yang diamati adalah dari spesies Turbo stenogyrus. Analisis pola pertumbuhan menunjukkan bahwa pola pertumbuhan T. stenogyrus adalah allometrik negatif. Amplifikasi DNA dengan gen COI menghasilkan panjang sekuen DNA 650 bp dengan hasil identifikasi sesuai dengan pengamatan morfologi yaitu Turbo stenogyrus. Berdasarkan analisis filogenetik dan jarak genetik, sekuen DNA yang dianalisis membentuk clade yang sama dengan spesies Turbo stenogyrus dengan jarak genetik terdekat 4,2%. Analisis genetik melalui gen COI sangat baik dan memberi kemudahan dalam identifikasi spesies gastropoda.
Shallot is an important commodity that becomes the basic food needs of the Indonesian people. Farmers generally use chemical or inorganic fertilizers in their cultivation activities. Inorganic fertilizers can accelerate the growing period because the nutrients can be absorbed directly by the soil. However, the continuous use of inorganic fertilizers can harm the soil and the environment, then to avoid this is to switch to using organic fertilizers. Manure is one of the organic fertilizers that can increase the availability of nutrients for plants. With the presence of intensive bacteria, it can accelerate the availability of nutrients. Bacteria whose role is to decompose organic matter are called PGPR. This study aims to (i) know the interactions (ii) know the single factor of cow manure (iii) know the single factor of bamboo root PGPR (iv) get the best interaction (v) get the best dose of cow manure (vi) get the best dose of bamboo root PGPR. This research was carried out in alluvial land in Tabalong District from March-June 2019. This study used a factorial randomized block design with the treatment is a dose of cow manure, namely: p1=10 t/ha, p2=20 t/ha, p3=30 t/ha, while the bamboo roots PGPR are: b1=20 ml, b2=25 ml, and b3=30 ml, so the combination is 9 treatments. The results showed that the observed variables were plant height, number of leaves, number of tubers, tuber diameter and tuber weight only affected plant height with a single factor treatment of cow manure and the best dose was 20 t/ha.Shallot is an important commodity that becomes the basic food needs of the Indonesian people. Environmental degradation and the utilization of excess are very influential in a reduction of thepotential of Turbinidae Gastropod Resources. Morphology of shells can changes due to adaptation tothe surrounding environment resulting in misidentification. This study aims to analyze morphology andmolecular character of Turbinidae. Morphometric analysis is done by comparing the measureddimensions of the shell, while the molecular identification is done by using the DNA barcoding usingCOI gene to species identification. The results showed a comparison of growth between thedimensions shell of the Lunella sp. showed the negative allometric and positive allometric. Characterof shell growth between dimensions of the Lunella sp. shows the negative allometric and positiveallometric. Molecular analysis identified Turbinidae was found Lunella sp. its similar to DNAsequence Lunella sp. Sulawesi. The molecular analysis identifies Turbinidae found is Lunella sp. withsimilarities of 98.41 and 98.31%. The environmental conditions can affect change the morphology andgenetic of Turbinidae. DNA barcoding has been successful to identify Turbinidae gastropods.
