Several sites of nuclear protein interaction within the promoter region of the Antirrhinum majus floral meristem identity gene SQUAMOSA were detected using an electrophoretic mobility shift assay. One of these sites displayed a particularly clear interaction with nuclear protein extracted from inflorescences but not with nuclear protein extracted from young, nonflowering plants. This site could thus represent a binding motif for a transcriptional activator. A South-western screen of an inflorescence cDNA expression library resulted in the isolation of several cDNAs representing two different genes named SBP1 and SBP2 (for SQUAMOSA-pROMOTER BINDING PROTEIN gene 1 and 2). Both genes encode highly similar protein domains which were found to be necessary and sufficient for binding DNA in a sequence-specific manner. This DNA-binding domain showed no similarity to known proteins in the databases. However, it is characteristic for a small family of gene products in A. majus and other plant species. Expression of SBP1 and 2 is developmentally regulated and their transcriptional activation precedes that of SQUAMOSA. The data presented support the idea that members of the newly identified SBP gene family function as transcription factors involved in the control of early flower development.
Anomalous flowering of the Antirrhinum majus mutant squamosa (squa) is characterized by excessive formation of bracts and the production of relatively few and often malformed or incomplete flowers. To study the function of squamosa in the commitment of an inflorescence lateral meristem to floral development, the gene was cloned and its genomic structure, a well as that of four mutant alleles, was determined. SQUA is a member of a family of transcription factors which contain the MADS‐box, a conserved DNA binding domain. In addition, we analysed the temporal and spatial expression pattern of the squa gene. Low transcriptional activity of squa is detectable in bracts and in the leaves immediately below the inflorescence. High squa transcript levels are seen in the inflorescence lateral meristems as soon as they are formed in the axils of bracts. Squa transcriptional activity persists through later stages of floral morphogenesis, with the exception of stamen differentiation. Although necessary for shaping a normal racemose inflorescence, the squa function is not absolutely essential for flower development. We discuss the function of the gene during flowering, its likely functional redundancy and its possible interaction with other genes participating in the genetic control of flower formation in Antirrhinum.
Aqueous solutions of a copolymer derivative of a polyacrylamide showed very interesting behavior, that in which the system evolves from one kind of double criticality (pressure-hypercritical point) to another (temperature-hypercritical point) as polymer molecular weight decreases. While in the neighboring region of the former point one expects a change from contraction to expansion upon mixing with increasing pressure; in the latter, mixing should be accompanied by a change in the sign of the excess enthalpy as temperature increases. L-L equilibria studies were performed in a wide range of (T, p) experimental conditions (300 < T/K < 460, 0 < p/bar < 700). Poly(N-isopropylacrylamide), usually called PNIPAAM, and its copolymer derivative poly(N-isopropylacrylamide/1-deoxy-1-methacrylamido-D-glucitol), herein referred to as CP, were investigated for several chain lengths and compositions. An He/Ne laser light scattering technique was used for the determination of cloud-point (T, p, x) conditions. The experimental results were used to assist in the determination of computed values at temperatures beyond experimental accessibility, which are obtained by the application of a modified Flory-Huggins model. The model also estimates the excess properties of these solutions. Because of the intrinsic selfassociating nature of these systems, all studied solutions show a lower critical solution temperature (LCST). Both modeling results and H/D isotope substitution effects suggest also the existence of upper critical solution temperatures (UCST) and therefore closed-loop-type phase diagrams. However, these upper-temperature branches are experimentally inaccessible. Pressure effects are particularly interesting. For a low-MW CP, experimental data display a tendency toward a reentrant T-p locus, which supports the conjecture that these systems are inherently of the closed-loop type. In the cases of PNIPAAMs and high-MW CPs, the T-p isopleths show extrema. The copolymer aqueous solutions under study in this work model a single chemical system where pressure-hypercritical behavior evolves to a temperaturehypercritical one as the chain length decreases.
