The objective was to assess the effect of adding various concentrations of dimethylformamide on characteristics of canine semen diluted in powdered coconut water (ACP-106C; ACP Biotecnologia, Fortaleza, CE, Brazil) and frozen at -196°C. Fifteen ejaculates were collected by manual stimulation from five adult Boxer dogs. The sperm-rich fraction was diluted in ACP-106C (ACP Biotecnologia) containing 10% egg yolk and divided into four aliquots. The cryoprotectants used for each aliquot were 6% glycerol (control group; CG) or 2%, 4%, or 6% dimethylformamide (DF2, DF4, and DF6, respectively). After thawing, total motility (mean ± SEM) for CG (58.4 ± 24.6) was higher (P < 0.05) than that of the other groups (2% dimethylformamide, 24.4 ± 12.3; 4% dimethylformamide, 26.5 ± 16.1; and 6% dimethylformamide, 21.7 ± 17.9). Furthermore, there was a greater percentage of fast, average, and slow moving sperm (assessed with computer-aided semen analysis; CASA) in CG in comparison with the other three groups. Therefore, based on concentrations tested in this study, dimethylformamide, together with ACP-106C (ACP Biotecnologia) and 10% egg yolk as a diluent, yielded unsatisfactory in vitro results for freezing canine semen.
Surveillance of visceral leishmaniasis in epidemiologically distinct locations in Juruti, a mining municipality in Pará State, Brazil Vigilância da leishmaniose visceral em localidades epidemiologicamente distintas em Juruti, um município minerário do Estado do Pará, Brasil Vigilancia de la leishmaniasis visceral en localidades epidemiológicamente diferentes en Juruti, municipio minero del Estado de Pará (Brasil) ABSTRACT Surveillance actions for human visceral leishmaniasis (HVL) were carried out in Juruti, a mining municipality in Pará State. A peri-urban (Santa Maria-SM) and a rural (Capiranga-CA) location were selected with or without HVL, respectively, for the execution of four biannual serologic inquiries (lysate ELISA) in canine populations (SM = 94, CA = 45) and three entomological surveys (CDC light traps, 18-6 h x4). Subsequently, the clinical status, as well as the infection by Leishmania, was investigated in 53 dogs (SM = 28; CA = 25) with parasitological (bone marrow/lymph, Giemsa), molecular (peripheral blood leukocytes, kDNA-PCR) and serological (ELISA) diagnoses assessing different antigens (lysate, k39, Hsp83 -screen test, ROC curve). Seroprevalence varied in SM (45; 40; 15; 15%) and in CA (22; 30; 8.5; 0%), presenting increasing average IgG rates in SM (320; 378; 951; 1866; p <0.05) despite the euthanasia of dogs after the second survey, and stable average IgG rates in CA (100; 159; 141; 0), where euthanasia was not conducted. The frequency rates of Lutzomyia longipalpis/Lutzomyia spp. differed in SM (279/296) and CA (4/6). Clinical and laboratory results were similar for dogs from SM and CA, respectively: infection (parasitological examination: 86 and 84%; kDNA-PCR: 100%), clinical status (asymptomatic: 43 and 56%; symptomatic: 57 and 44%), and specificity by ELISA (100%). On the other hand, sensitivity (lysate: 44 and 18%; Hsp83: 48 and 27%; k39: 48 and 41%) and IgG levels (£ 6,400; £ 200) varied, respectively. The profile of canine infection in localities with or without HVL transmission differed only in terms of the level/evolution of IgG, which makes the temporality of investigations necessary, especially in quiet and isolated areas that present a low vector density and where the euthanasia of dogs would become unnecessary. The best serological test was ELISA-k39.
RESUMO O objetivo deste trabalho foi caracterizar o manejo sanitário e descrever as características da ovinocaprinocultura na microrregião homogênea (MRH) de Teresina, Piauí, Brasil. Foram investigadas 45 propriedades capriovinícolas, distribuídas em 14 municípios. O número médio de animais foi de 129 e 121 nos rebanhos caprinos e ovinos, respectivamente. Os animais utilizados na formação dos rebanhos base tiveram como origem os estados da Bahia, Pernambuco e Ceará. Animais SPRD (45,8%) e da raça Santa Inês (35,4%) são os mais encontrados entre os ovinos e na espécie caprina animais SPRD (55,3%) e animais da raça Anglo-Nubiana (28,6%). O estudo demostrou que 42,2% propriedades pesquisadas exploravam bovinos e caprinos e/ou ovinos. O sistema de criação extensivo foi adotado em 71,1% dos criatórios. As práticas sanitárias adotadas com maior frequência foram descarte de agulhas após o uso, isolamento de animais doentes e corte e desinfecção do umbigo. A desverminação foi a prática utilizada no controle de verminoses por 100,0% dos produtores. A vacinação foi adotada em 48,8% dos rebanhos. As alterações clínicas mais citadas foram linfadenite caseosa (64,4%), pododermatite (62,2%) e diarreia (60,0%). Portanto, a ovinocaprinocultura na MRH de Teresina é praticada, em sua grande maioria, de forma extensiva e que, apesar de contar com instalações razoáveis, ainda apresenta deficiências quanto à utilização de práticas de manejo sanitário, o que acaba impossibilitando a prevenção e o controle de doenças, principalmente as de origem infecciosas e parasitárias.
