Commercial probiotics have bacteria which offer scope for specific treatment as a probiotic against diarrhoea, boosting immune response and relieving stress. Research in probiotics and aquaculture has been done to promote sustainable aquaculture. Studies on microbial ecology in aquaculture and their benefits on human systems are to be assessed. Hence, the present study was aimed at the isolation and characterization of these bacteria obtained from various commercial probiotics. The commercial probiotic samples were collected for isolation using MRS agar. The colonies were selected on the basis of colonial morphology. Isolates were put through cell morphology, physiology and different biochemical tests. Probiotic sample confirmation was done using 16srRNA molecular technique. MEGA7 was used to conduct phylogenetic evolutionary analysis and tabulate a distance matrix. In results, the isolates manifested non-identical growth patterns at dissimilar conc. of NaCl (2.0,4.0 and 6.5) oxygen and at different temperatures (150C, 300C and 450C). On the basis of sugar utilization, physiological testing, biochemical characterization and genetic identification tests, all isolates were established to different species of Lactobacillus rhamnosus, Pediococcus acidilactici and Lactobacillus Plantarum. Pongamia pinnata L, Larvicidal, Mosquito vectors, Culex quinquefasciatus, Aedes aegypti, Anopheles stephensi Objective: To evaluate the mosquito larvicidal activity of Pongamia pinnata (P. pinnata) extracts against three mosquito vectors. Methods: The methanol and hydroalcohol extracts of bark part of P. pinnata L were tested against fourth instar larvae of Culex quinquefasciatus, Aedes aegypti and Anopheles stephensi. The mortality was observed 24 h and 48 h after treatment, data was subjected to probit analysis to determine lethal concentration (LC 50 and LC 90) to kill 50 and 90 percent of treated larvae of tested species. Results: The larval mortality was found in both methanol and hydroalcohol extracts of P. pinnata against Culex quinquefasciatus, Aedes aegypti and Anopheles stephensi with LC 50 values of 84.8, 118.2 and 151.7 ppm; 97.7, 128.3 and 513 ppm. The highest larval mortality was found in methanol extract of P. pinnata when comparable to the hydroalcohol extract. Conclusions: These results suggest that both methanol and hyrdoalcohol extracts have the potential to be used as an ideal ecofriendly approach for the control of disease vectors. This could lead to isolation of novel natural larvicidal compounds.
Vibrio parahaemolyticus is the most common species among crustaceans, often causing various diseases and significant losses in aquaculture. Acute hepatopancreatic necrosis disease (AHPND) is a newly emerging shrimp disease that has severely damaged the global shrimp industry. This species of bacteria is associated with gastrointestinal illness in humans and has been implicated in foodborne disease. The present study carried out, isolation and characterization of pathogenic bacterial flora isolated from the infected hepatopancreas of vannamei, obtained from various aquafarms in Andhra Pradesh, India, on 11th June 2018. The collected samples were plated on TCBS- (Thiosulfate-Citrate-Bile salt-Sucrose) agar medium and Hi -Chrome vibrio, as described in Bergey's manual of systematic bacteriology. Isolated colonies were subjected to the following tests- microscopic examination, growth at different temperatures, growth at different NaCl concentrations, and biochemical tests. Further purity, maintenance, and propagation of purified cultures were done. The microbial culture was identified using 16s rRNA molecular technique. Phylogenetic Evolutionary analyses and distance matrix were conducted in MEGA7.In the present study, different samples were screened, a total of three green colonies (V44, V45, V46) were isolated, identified by biochemical tests and genetic identification as Vibrio parahaemolyticus. There is increasing scientific evidence and commercial interest for using probiotics for eliminating and handling of specific diseases. Probiotics can be evaluated for its role and performance against isolated pathogens from contaminating sources. The present work reports on invitro antimicrobial activity of commercial selected probiotics against pathogenic microbe Vibrio parahaemolyticus. The work also describes cytotoxic activities using MTT assay and adherence studies of selected probiotics. Results for the studies showed maximum zone of inhibition 13.66±0.46mm in probiotic enteroplus,12.33±0.93mm in lactobacillus (NCIM2056) and 10.66±0.93mm in Avant Bact. Cytotoxicity was expressed as IC50(µg/ml) values, observed on CaCO cell lines for different probiotics. Avant Bact showed a IC50 value of 104.7745, Lactobacillus (NCIM2056) a value of 58.13223 and Enteroplus a value of 50.09716. These values expressed different safety aspects of probiotics used for study. Finally the adherence study was done to check probiotic colonizing capacity. The probiotics showed varied adherence capacity against caco cell lines. Enteroplus has % adhesion of 10.25±0.74, Avant Bact. 7.25±0.82 and Lactobacillus (NCIM2056) 7.5±1.12. In conclusion antimicrobial results show importance of probiotics to be used against specific gastro intestinal diseases. Cytotoxicity determines safety aspects of probiotics and adherence study determines probiotic as a promising candidate for in vivo studies.
