The effect of temperature, moisture content and pH during solid-state fermentation (SSF) of MD2 pineapple peel with Rhizopus oryzae (MUCL 28168) was evaluated on the release of bioactive compounds with antioxidant capacity. Applying a central composite design, it was found that temperature had a significant effect (p < 0.05) on the total phenolic content and DPPH antioxidant activity while for the ABTS radical elimination activity, the factor that presented a significant effect was the pH (p < 0.05); as this factor increases, the antioxidant activity enhances. The optimal conditions for fermentation process were 80% of moisture content, pH 5.5, temperature 37.3 °C and 24 h of process to maximize phenolic content and antioxidant activity. Gallic acid, chlorogenic acid, caffeic acid and cinnamic acid were identified in the extracts by HPLC analysis. These results permit to conclude that SSF of pineapple peel is an effective bioprocess for the release of phenolic compounds with antioxidant activity. Graphical abstract
Abstract. Leaf gas-exchange models show considerable promise as paleo-CO2 proxies. They are largely mechanistic in nature, provide well-constrained estimates even when CO2 is high, and can be applied to most subaerial, stomata-bearing leaves from C3 taxa, regardless of age or taxonomy. Here we place additional observational and theoretical constraints on one of these models, the Franks model. In order to gauge the model's general accuracy in a way that is appropriate for fossil studies, we estimated CO2 from 40 species of extant angiosperms, conifers, and ferns based only on measurements that can be made directly from fossils (leaf δ13C and stomatal density and size) and a limited sample size (1–3 leaves per species). The mean error rate is 28 %, which is similar to or better than the accuracy of other leading paleo-CO2 proxies. We find that leaf temperature and photorespiration do not strongly affect estimated CO2, although more work is warranted on the possible influence of O2 concentration on photorespiration. Leaves from the lowermost 1–2 m of closed-canopy forests should not be used because the local air δ13C value is lower than the global well-mixed value. Such leaves are not common in the fossil record, but can be identified by morphological and isotopic means.
Antioxidant phenolic compounds were extracted from fermented samples of Golden pineapple peels via an ultrasound method. The fermentation conditions to maximize the production of phenolic content and antioxidant activity were previously determined (pH: 5.5, T: 37.3 °C and 85% moisture content). A central composite design with 20 treatments was applied to evaluate the effect of the ethanol concentration, time, and temperature on the production of phenolic compounds and antioxidant activity of the extracts. The statistical analysis showed that the optimal conditions to produce extracts with high phenolic content and antioxidant activity were: 62 °C, 30 min and 58% ethanol. We obtained 866.26 mg gallic acid equivalents (GAE)/g d.m in total phenolic content and for antioxidant activity expressed as percentage inhibition, 80.06 ± 1.02% for ABTS and 63.53 ± 2.02% for DPPH, respectively. The bioactive compound profile in the extracts was identified and quantified using ultra-high performance liquid chromatography (UHPLC), this method showed the presence of rosmarinic acid, caffeic acid, vanillic acid, p-coumaric acid, ferulic acid, quercetin-3 glucoside, rutine, quercetin, kaempherol-3 glucoside and gallic acid, demonstrating the great potential of these by-products to obtain components that can benefit the consumer’s health.
La fermentación del cacao es determinada por el crecimiento de sucesiones de diversos grupos microbianos que varían de una región a otra. En el presente estudio se realizó una caracterización de las muestras tomadas de fermentaciones de unidades productivas del Valle del Cauca a las cuales se les midió la concentración de levaduras, bacterias lácticas, bacterias acéticas y aerobios mesófilos durante 5 días. Adicionalmente se determinó la concentración de azúcares reductores y pH tanto del cotiledón como de la testa mucilaginosa. Finalizada la fermentación se determinó el grado de fermentación (prueba de corte e índice de fermentación). Se presentó un crecimiento de levaduras, bacterias acéticas y bacterias lácticas constante durante los 5 días de fermentación evaluados, a excepción del muestreo 1 que presentó un descenso en la concentración de BAA. Se presentaron temperaturas promedio de fermentación de 26, 27 y 25 °C, consumo de azúcares fue de 64.40, 88.78 y 98.27% para los puntos de muestreo 1, 2 y 3, respectivamente. Conocer el comportamiento de los grupos microbianos y sus respuestas bioquímicas, permiten entender las relaciones entre el crecimiento de la microbiota del cacao y su calidad final, que a futuro permitan mejorar la calidad de los productos derivados.
