RESUMOErythrina crista-galli L. (FABACEAE), corticeira-do-banhado, é uma árvore nativa do Brasil, Uruguai, Paraguai, Bolívia e Argentina. As Regras de Análises de Sementes -RAS não possuem orientações para a realização de testes de germinação para esta espécie. O objetivo deste trabalho foi testar o desempenho de tratamentos para a superação de dormência das sementes e recomendar condições adequadas de substrato, temperatura e fotoperíodo para a realização de testes de germinação. Foi utilizado o delineamento inteiramente casualizado utilizando-se o software SASM-Agri® para a análise de variância e a comparação de médias pelo teste de Tukey, a 5% de probabilidade de erro. O tratamento indicado para a superação da dormência foi a escarificação mecânica e a temperatura ótima para os testes de germinação de 30ºC constantes. Não houve diferença entre tratamentos em relação aos substratos testados nem quanto aos fotoperíodos, indicando-se assim o substrato entre papel e com 8h de luz por sua praticidade e economia.

ABSTRACTErythrina crista-galli L. (FABACEAE) is a tree native to Brazil, Uruguay, Paraguay, Bolivia and Argentina. Seeds of test requirements does not have guidance for conducting tests of germination for the species. The aim of this study was to test the performance of treatments to overcome seed dormancy and recommend appropriate conditions of substrate, temperature and photoperiod to germination tests. We used a completely randomized design using the SASM-Agri® software for the analysis of variance and comparison of means by Tukey test, at 5% probability of error. The treatment to overcome dormancy was mechanical scarification and the optimum temperature to germination tests was 30°C constant. There was no difference between treatments in relation to the substrates tested or as the photoperiod is indicating the substrate between paper and 8h light for its practicality and economy.
Erythrina crista-galli is a native tree from South America with an important role on pharmaceutical studies. The objective of this study was to model the ancient and current ecological niches distribution of E. crista-galli in South America, contributing to the discussion about species management and conservation. A reduction in the potential area of species occurrence was detected by comparing past and present distribution. Based on the obtained results, it is expected that this species tends to expand its frontiers if the natural dynamic of the extant populations is guaranteed in southern South America. Therefore, management and conservation of E. crista-galli should focus on allowing the expansion of native forest formations where this species occurs. Additionally, further studies on the medicinal properties of the species may valorize and promote a higher interest in rational exploitation and conservation of E. crista-galli in its natural environment.

(FABACEAE) é uma árvore nativa do Brasil, Uruguai, Paraguai, leste da Bolívia e Argentina, podendo ser utilizada na arborização e na restauração de ambientes naturais. Não foram encontradas na literatura metodologias para o isolamento de DNA genômico da espécie. O objetivo do trabalho foi testar três protocolos de isolamento de DNA aplicados a folhas secas à temperatura ambiente verificando a quantidade e qualidade do DNA isolado. Folhas secas mostraram constituir boa fonte de material para as avaliações genéticas da espécie estudada, entretanto, novos testes são necessários no sentido de aperfeiçoar os protocolos e solucionar as contaminações por polissacarídeos e polifenóis nas amostras.

Dried leaves showed constitute good source of genetic material, however, new tests are needed to improve the protocols to solve contamination by polysaccharides and polyphenols in samples.
