A plasma needle using a dielectric barrier discharge reactor at atmospheric pressure with a funnel-shaped nozzle was developed. The preliminary characteristics of the plume and applications to the ablation of cultured human hepatocellular carcinoma (HCC) BEL-7402 cell line were presented. The effect of oxygen, which was injected into argon plasma afterglow region through a steel tube, was studied. The efficiency of argon-oxygen plasma depends sensitively on the oxygen concentration in argon plasma. Large differences between spectra in atmosphere and those in Dulbecco’s modified eagle medium are found. It is found that ultraviolet rays, O, OH, and Ar radicals can reach the bottom of solution and act on HCC cells and there is an optimum input power to get the most radicals.
ObjectiveTo investigate the significance of the dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) parameters of diffuse spinal bone marrow infiltration in patients with hematological malignancies.Materials and MethodsDynamic gadolinium-enhanced MR imaging of the lumbar spine was performed in 26 patients with histologically proven diffuse bone marrow infiltration, including multiple myeloma (n = 6), acute lymphoblastic leukemia (n = 6), acute myeloid leukemia (n = 5), chronic myeloid leukemia (n = 7), and non-Hodgkin lymphoma (n = 2). Twenty subjects whose spinal MRI was normal, made up the control group. Peak enhancement percentage (Emax), enhancement slope (ES), and time to peak (TTP) were determined from a time-intensity curve (TIC) of lumbar vertebral bone marrow. A comparison between baseline and follow-up MR images and its histological correlation were evaluated in 10 patients. The infiltration grade of hematopoietic marrow with plasma cells was evaluated by a histological assessment of bone marrow.ResultsDifferences in Emax, ES, and TTP values between the control group and the patients with diffuse bone marrow infiltration were significant (t = -11.51, -9.81 and 3.91, respectively, p < 0.01). Emax, ES, and TTP values were significantly different between bone marrow infiltration groups Grade 1 and Grade 2 (Z = -2.72, -2.24 and -2.89 respectively, p < 0.05). Emax, ES and TTP values were not significantly different between bone marrow infiltration groups Grade 2 and Grade 3 (Z = -1.57, -1.82 and -1.58 respectively, p > 0.05). A positive correlation was found between Emax, ES values and the histological grade of bone marrow infiltration (r = 0.86 and 0.84 respectively, p < 0.01). A negative correlation was found between the TTP values and bone marrow infiltration histological grade (r = -0.54, p < 0.01). A decrease in the Emax and ES values was observed with increased TTP values after treatment in all of the 10 patients who responded to treatment (t = -7.92, -4.55, and 5.12, respectively, p < 0.01).ConclusionDCE-MRI of spine can be a useful tool in detecting diffuse marrow infiltration of hematological malignancies, while its parameters including Emax, ES, and TTP can reflect the malignancies' histological grade.
Our previous studies demonstrated that LAPTM4B-35 is overexpressed in a variety of solid cancers including hepatocellular carcinoma (HCC), and is an independent factor for prognosis. LAPTM4B-35 overexpression causes carcinogenesis and enhances cancer growth, metastasis and multidrug resistance, and thus may be a candidate for therapeutic targeting. The present study shows ethylglyoxal bisthiosemicarbazon (ETS) has effective anticancer activity through LAPTM4B-35 targeting. Bel-7402 and HepG2 cell lines from human HCC were used as cell models in which LAPTM4B-35 is highly expressed, and a human fetal liver cell line was used as a control. The results showed ETS has a specific and pronounced lethal effect on HCC cells, but not on fetal liver cells in culture. ETS also attenuated growth and metastasis of human HCC xenograft in nude mice, and extended the life span of mice with HCC. ETS induced HCC cell apoptosis, and upregulated a large number of proapoptotic genes and downregulated antiapoptotic genes. When endogenous overexpression of LAPTM4B-35 was knocked down with RNAi, the killing effect of ETS on HepG2 cells was significantly attenuated. ETS also inhibited phosphorylation of LAPTM4B-35 Tyr285, which involves in activation of the PI3K/Akt signaling pathway induced by LAPTM4B-35 overexpression. In addition, the induction of alterations in quantity of c-Myc, Bcl-2, Bax, cyclinD1 and Akt-p molecules in HepG2 cells by LAPTM4B-35 overexpression could be reversed by ETS. Conclusion: ETS is a promising candidate for treatment of HCC through LAPTM4B-35 protein targeting.
The positioning technology to find shallow underground vibration sources based on a wireless sensor network is receiving great interest in the field of underground position measurements. The slow peaking and strong multi-waveform aliasing typical of the underground vibration signal result in a low extraction accuracy of the time difference and a poor source-positioning accuracy. At the same time, the transmission of large amounts of sensor data and the host computer’s slow data processing speed make locating a source a slow process. To address the above problems, this paper proposes a method for high-precision time-difference measurements in near-field blasting and a method for its hardware implementation. First, based on the broadband that is typical of blast waves, the peak frequency of the P-wave was obtained in the time–frequency domain, taking advantage of the difference in the propagation speed of the P-wave, S-wave, and the surface wave. Second, the phase difference between two sensor nodes was found by means of a spectral decomposition and a correlation measurement. Third, the phase ambiguity was eliminated using the time interval of the first break and the dynamic characteristics of the sensors. Finally, following a top-down design idea, the hardware system was designed using Field Programmable Gate Array(FPGA). Verification, using both numerical simulations and experiments, suggested that compared with generalized cross-correlation-based time-difference measurement methods, the proposed method produced a higher time-difference resolution and accuracy. Compared with the traditional host computer post-position positioning method, the proposed method was significantly quicker. It can be seen that the proposed method provides a new solution for solving high-precision and quick source-location problems, and affords a technical means for developing high-speed, real-time source-location instruments.
