It wasfound that irradiation of aqueous solution of aluminium sulphophthalocyanine (SPcAl) by light with wavelength of 805 nm in the presence of sodium ascorbate results in photodynamic type I effect. S2PcA1 in adsorbed state on proteins or polysaccharide structures was shown to possess rather intense long wavelength absorption band in the NIR spectral region. The comparative tests of SPcAl with sodium ascorbate on mice with Ehrlich carcinoma and leucosis P-388 showed that photodynamic efficiency in the case of NIR irradiation (790 nm) is not lower than using visible irradiation (675 nm).
.ABSTRACT.It is very important to develop new sensitizer which could be efficiently excited at wavelengths exceeding 685 nm where own tissue absorption can be neglected. We have synthesize water soluble phthalocyanine derivative having phosphonate groups as substituents in macrocycle. New sensitizer efficiently absorbs in spectral range 685 -710 nm. It exhibits intense fluorescence in range 690 -720 nm. The photodynamic activity of new sensitizer is higher than in the case of sulfonated aluminium phthalocyanine.
BACKGROUND.A wavelength of photosensitizer absorption maximum and, correspondingly, of exciting laser source is one of the most important parameters determing the depth of photodynamic action on tissue. The widely used now haematoporphyrin derivatives (such as "Photofrin", "Photosan", "Photohem")'-3 provide only superficial action (< 4 -S mm) because of strong absorption of unsensitized tissues in spectral range 625 -635 n.m.The widely investigated now phthalocyanine (such as, for example, sulfonated phthalocyanine aluminium4 -PcSA1 ) have in this respect doubtless advantages over haematoporphyrin derivatives. Owing to red shifted PcSA1 long wavelength absorption maximum, the contribution of photosensitizer in absorbance of sensitized tissue begin to predominate offer it's own absorption (Fig. 1). Therefore, it is very important to develop new phthalocyanine derivatives which could be efficiently excited at wavelength exceeding 685 nm where own tissue absorption can be neglected. Fig. 1.
