In molecular and cellular biological research, cell isolation and sorting are required for accurate investigation of a specific cell types. By employing unique cell properties to distinguish between cell types, rapid and accurate sorting with high efficiency is possible. Though conventional methods can provide high efficiency sorting using the specific properties of cell, microfluidics systems pave the way to utilize multiple cell properties in a single pass. This improves the selectivity of target cells from multiple cell types with increased purity and recovery rate while maintaining higher throughput comparable to conventional systems. This review covers the breadth of microfluidic platforms for isolation of cellular subtypes based on their intrinsic (e.g., electrical, magnetic, and compressibility) and extrinsic properties (e.g., size, shape, morphology and surface markers). The review concludes by highlighting the advantages and limitations of the reviewed techniques which then suggests future research directions. Addressing these challenges will lead to improved purity, throughput, viability and recovery of cells and be an enabler for novel downstream analysis of cells.
The present review provides information relevant to issues and challenges in MEMS testing techniques that are implemented to analyze the microelectromechanical systems (MEMS) behavior for specific application and operating conditions. MEMS devices are more complex and extremely diverse due to the immersion of multidomains. Their failure modes are distinctive under different circumstances. Therefore, testing of these systems at device level as well as at mass production level, that is, parallel testing, is becoming very challenging as compared to the IC test, because MEMS respond to electrical, physical, chemical, and optical stimuli. Currently, test systems developed for MEMS devices have to be customized due to their nondeterministic behavior and complexity. The accurate measurement of test systems for MEMS is difficult to quantify in the production phase. The complexity of the device to be tested required maturity in the test technique which increases the cost of test development; this practice is directly imposed on the device cost. This factor causes a delay in time-to-market.
Master mold fabricated using micro milling is an easy way to develop the polydimethylsiloxane (PDMS) based microfluidic device. Achieving high-quality micro-milled surface is important for excellent bonding strength between PDMS and glass slide. The aim of our experiment is to study the optimal cutting parameters for micro milling an aluminum mold insert for the production of a fine resolution microstructure with the minimum surface roughness using conventional computer numerical control (CNC) machine systems; we also aim to measure the bonding strength of PDMS with different surface roughnesses. Response surface methodology was employed to optimize the cutting parameters in order to obtain high surface smoothness. The cutting parameters were demonstrated with the following combinations: 20,000 rpm spindle speed, 50 mm/min feed rate, depth of cut 5 µm with tool size 200 µm or less; this gives a fine resolution microstructure with the minimum surface roughness and strong bonding strength between PDMS–PDMS and PDMS–glass.
